摘  要:目的:探讨胎盘组织中ZNF331mRNA及其蛋白表达在早发型重度子痫前期发病中的作用。方法:采用RTPCR、Western blot方法检测50例正常晚期妊娠者和20例早发型重度子痫前期、28例晚发型重度子痫前期患者胎盘组织中ZNF331及其蛋白的表达水平。结果:正常晚期妊娠组胎盘组织中ZNF331mRNA表达量为1.08±0.16,早发型重度子痫前期组为1.91±0.31,晚发型组为1.13±0.27。其中早发型重度组ZNF331的表达显著高于晚发型组和正常组,而晚发型组与正常组无统计学差异(P>0.05)。ZNF331蛋白在早发型重度子痫前期患者胎盘组织中的表达量为(59.00±5.32)μg/100μg总蛋白,显著高于晚发型重度子痫前期患者和正常妊娠孕妇,晚发型组与正常妊娠组无明显差异。ZNF331mRNA及其蛋白表达与新生儿体重呈显著负相关(r=-0.798,P=0.000;r=-0.874,P=0.000)。随着新生儿体重的降低,ZNF331表达水平显著升高(P<0.05)。结论:早发型重度子痫前期患者胎盘组织ZNF331及其蛋白表达明显高于晚发型组和正常晚期妊娠组,提示其异常表达可能是早发型重度子痫前期的发病原因,可能与滋养细胞浸润能力下降有关,并且与新生儿体重及预后密切相关。Objective: To explore the roles of placental ZNF331 mRNA and its protein in pathogenesis of early- onset severe pre- eclampsia. Methods: RT -PCR and Western blot were used to detect expression levels of ZNF331 mRNA and its protein in placental speci- mens of 50 normal late pregnant women, 20 patients with early - onset severe preeclampsia and 28 patients with late - onset severe pre- eclampsia. Results: The expression levels of ZNF331 mRNA in placental tissue of normal late pregnant women, patients with early- onset severe preeclampsia and patients with late - onset severe preeclampsia were ( 1.08 ± 0. 16 ), ( t. 91 ± 0. 31 ) and ( 1.13 ± 0. 27 ), respectively; the expression level of ZNF331 mRNA in placental tissue of patients with early - onset severe preeclampsia was significantly higher than those of patients with late - onset severe preeclampsia and normal late pregnant women, but there was no statistically significant differ- ence between patients with late - onset severe preeclampsia and normal late pregnant women ( P 〉 0. 05 ) ; the expression level of ZNF33 pro- tein in placental tissue of patients with early- onset severe preeclampsia was (59. 00 ± 5. 32 )μg/100μg total protein, which was signifi- candy higher than those of patients with late - onset severe preeclampsia and normal late pregnant women, but there was no statistically sig- nificant difference between patients with late -onset severe preeclampsia and normal late pregnant women; there was a significantly negative correlation between expression levels of ZNF331 mRNA and its protein and neonatal weight ( r = - 0. 798, P = 0. 000 ; r = - 0. 874, P = 0. 000) ; the expression level of ZNF331 increased significantly with the decrease of neonatal weight (P 〈 0. 05 ) . Conclusion: The expres- sion levels of ZNF331 mRNA and its protein in placental tissue of patients with early - onset severe preeelampsia was significantly higher than those of patients with late -onset severe preeclampsia and normal late
