摘  要:为了研究转基因741杨对美国白蛾抗虫稳定性及其生长发育的影响,选择转基因741杨Pb29、Pb17两个无性系为试验材料,测定各龄幼虫死亡率、各龄虫体重、排粪量。试虫取食转基因741杨2周后,测定其中肠淀粉酶和蛋白酶活性。结果表明,转抗虫基因杨树饲养的美国白蛾幼虫死亡率均高于CK 5.34%,高抗Pb29饲养的幼虫死亡率为93.84%,中抗Pb17的为50.01%;存活幼虫发育不正常,发育历期明显延迟,体重增加速率和排粪量均显著降低,高抗Pb29和中抗Pb17饲养的虫体重增长率分别为CK的3.8%和36%,排粪量分别比CK少0.585 7g和0.310 5g;取食转基因无性系的幼虫中肠蛋白酶及淀粉酶活性显著降低,Pb29对其蛋白酶和淀粉酶的抑制率分别为77.93%和96.38%,Pb17的抑制率分别为36.31%和21.14%。因此,转基因741杨树对美国白蛾幼虫中肠酶的抑制作用与对其生长发育的阻滞有良好的相关性,转基因741杨无性系对美国白蛾幼虫仍具有稳定抗性。Using leaves with different insect resistance froms clones Pb29、Pbl7of transgenic hybrid poplar 741 [Populus alba L.×(P.davidiana Dode+ P.simonii Carr.)× P.tomentosa Carr.]as experimental materials and the normal poplar 741as CK to feed the larvae of Hyphantria cunea,the stability of insect-resistant transgenic poplar 741to Hyphantria cunea and the effects on the growth and develoment of Hyphantria cunea were studied in this research,determing the mortality of different instar larvae and the weight of the worm and frass.Two weeks after Larvae were fed with transgenic poplar 741,measuring the activities of midgut amylase and protease were measured.The results indicated that the mortality of Hyphantria cuneafeeding on transgenic poplar 741was higher than control 5.34%,larval mortality feeding on clones Pb29and Pb17was 93.84% and 50.01%.The survival larvae had no normal development and therefore the development period was postponed;the growth speed,the weight increasing rate and the feces amount were decreased.The weight growth rates of insects feeding on clones Pb29and Pb17account for 3.8%and 36% of CK,respectively,the amount of feces was less 0.585 7g and 0.310 5g than CK.Aftler the insects were fed with transgenic clones,the activity of midgut enzymes(toxic proteins amylase)significantly reduced.The results also showed that the activity of the midgut proteases and amylases was also decreased.The inhibition rate of proteases and amylases of insects after feeding with Pb29was about 77.93%and 96.38%,while after that feeding with Pb17,the inhibition rate was 36.31% and 21.14%.The decrease on activities of the midgut proteases showed good correlation with the retardation on the growth of the larvae fed with transgenic poplar 741,which still has a stable resistance to the Hyphantria cunea.
