摘  要:目的探讨去乙酰化酶沉默信息调节因子2相关酶1(SIRT1)与大鼠睡眠呼吸暂停指数的相关性及不同缺氧模型对大鼠低位脑干SIRT1表达量的影响。方法对健康SD大鼠33只进行睡眠呼吸监测,随后取其低位脑干组织,应用Western blot对低位脑干组织SIRT1相对表达量进行检测,分析大鼠睡眠呼吸暂停指数与SIRT1表达量的相关性。另取健康sD大鼠36只按随机数字表法分为4组:对照组、持续缺氧组(CH组)、间歇缺氧组(IH组)和间歇缺氧伴高二氧化碳组(IHH组),每组9只,使用Western blot法检测其低位脑干组织SIRT1相对表达量,比较4组大鼠SIRT1表达量的差异。结果SIRT1与快动眼睡眠期自发性睡眠呼吸暂停指数(SPAI)呈负相关(r=-0.617,P〈0.01),与REM期叹息后睡眠呼吸暂停指数(PSAI)呈正相关(r=0.535,P〈0.01)。对照组、CH组、IH组、IHH组脑干SIRT1蛋白相对表达量分别为1.4±1.0、1.4±0.6、2.5±1.0和1.6±1.2,差异有统计学意义(F=2.964,P〈0.05)。结论脑干SIRT1表达量低的大鼠可能因抗氧化应激能力低,中枢调控不稳,导致其自发呼吸暂停增多。缺氧情况下SIRT1表达升高,推测是一种保护机制。Objective To explore the correlation between SIRT1 expression in brainstem and apnea hypopnea index and the effect of hypoxia on SIRTl expression in rats. Methods Thirty-three healthy Sprague-Dawley(SD) rats underwent sleep monitoring and SIRTl protein in brainstems was measured by Western blot. The correlation of sleep apnea index and SIRT1 protein level in brainstem was analysed. Another 36 adult male SD rats were randomly and equally divided into 4 groups : normal control ( NC ) group, continuous hypoxia (CH) group, intermittent hypoxia (IH) group and intermittent hypoxia with hypercapnia (IHH) group. The expression of SIRT1 protein of the lower braiustems of the 4 groups were measured by Western blot. Results The level of SIRT1 in the brainstem was negtively correlated with the frequency of spontaneous sleep apnea in REM ( r = - 0. 617,P 〈 0. 001 ) while positively correlated with the frequency of post sigh sleep apnea in REM ( r = 0. 535,P 〈 0. 01 ). The relative expression level of SIRT1 in NC, CH, IH, IHH group were(1.4±1.0),(1.4±0.6),(2.5 ±1.0),(1.6±1.2),respectively. There was a significant difference in the SIRT1 level among groups ( F = 2. 964, P 〈 0. 05 ). Conclusions The rats with lower level of SIRT1 may have lower anti-oxidative stress capability which results in higher frequency of spontaneous sleep apnea. Under the circumstance of hypoxia, the fact that the expression of SIRT1 protein was elevated may be a protective mechanism.
