摘 要:目的:研究非酶糖基化终末产物受体(RAGE)在大鼠睾丸Leydig细胞上的表达及非酶糖基化终末产物(AGEs)对大鼠睾丸Leydig细胞睾酮合成的抑制作用。方法:原代培养大鼠睾丸Leydig细胞,RT-PCR和免疫荧光技术检测RACE在大鼠Leydig细胞上表达,不同浓度AGEs处理Leydig细胞(25、50、100、200μg/ml),ELISA法测定睾酮分泌量。结果:RT-PCR和免疫荧光结果表明RAGE在大鼠睾丸Leydig细胞上表达,不同浓度AGEs处理后,人绒毛膜促性腺激素(hCG)诱导的Leydig细胞睾酮合成量呈剂量浓度依赖性下降,与对照组相比,50、100、200μg/ml AGEs处理组差异显著(P<0.01)。结论:大鼠Leydig细胞上存在RAGE受体,AGEs显著抑制原代培养大鼠Leydig细胞睾酮的分泌。Objective : To study the expression of the receptor for advanced glycation end products (RAGE) and the inhibitory effect of advanced glycation end products (AGEs) on testosterone production in rat Leydig ceils. Methods : Rat Leydig cells were primarily cultured and the expression of RAGE in the Leydig cells was detected by RT-PCR and immunofluorescence staining. The Leydig cells were treated with AGEs at the concentrations of 25, 50, 100 and 200 μg/ml, respectively, and the testosterone content was determined by ELISA. Results: RT-PCR and immunofluorescence staining exhibited the expression of RAGE in the rat Leydig cells. AGEs remarkably suppressed hCG-induced testosterone production in the Leydig cells in a concentration-dependent manner in the 50, 100 and 200 μg/ml groups as compared with the control (P 〈 0.01 ). Conclusion : RAGE exists in rat Leydig cells and AGEs can significantly inhibit the secretion of testosterone in primarily cultured rat Leydig cells. Nutl J Androl, 2014, 20 (5) : 410 -413
