摘 要:目的 :建立测定阿魏酸衍生物(BL)含量及其有关物质的高效液相色谱法。方法 :采用Agilent Eclipse C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.1%乙酸(42:58),流速1.0mL·min-1;检测波长228nm;进样量20μL;柱温:40℃。结果 :在该条件下BL主峰与有关物质得到很好的分离;检测浓度在0.08~0.32mg·mL-1范围内具有良好的线性关系(r=0.9996);检测限和定量限分别为8ng和36ng。结论 :该方法准确可靠,灵敏度高,专属性好,可用于BL含量及其有关物质的测定。Objective: To establish an RP-HPLC method for determination of the content of ferulic acid derivative (BL) and its related substances. Methods: Agilent Eclipse C188(250mm×4.6mm,5μm) column was adopted;the mobile phase consisted of methanol-0.1% acetic acid (42:58) at the flow rate of 1.0 mL.min-1;The ultraviolet detection wavelength was set at 228nm;Loaded volume was 20μL;the column temperature was 40℃. Results: Using this method,BL and its related substances were well separated;The standard curve was rectilinear in the range of 0.08mg-mL-1 - 0.32 mg.mL-t (r=0.9996) ;The limit of detection and quantification were 8ng and 36ng. Conclusion: The method appeared to be accurate, reliable, sensitive, specific and can be used for the determination of the content BL and its related substances.
