狗牙根黑穗病病原菌的鉴定及其培养特性

摘  要:狗牙根(Cynodon dactylon)是耕地、果园及其他草种草坪常见的恶性杂草,为探索其生物防治方法,试验从患黑穗病狗牙根穗部分离到一株真菌LHL1,对其进行分子鉴定及生物学特性研究。液体发酵培养确定其最佳碳源和氮源及最适pH;苯胺蓝染色、光学显微镜镜检法观察感病狗牙根各部位菌丝和孢子分布情况。结果表明,菌株LHL1为担子菌亚门(Basidiomycotina)黑粉菌目(Ustilaginales)黑粉菌属(Ustilago);该菌的最佳碳源和氮源分别为甘油和水解乳蛋白,最佳pH为5;镜检结果显示在感病植株的叶鞘内表皮有大量菌丝及孢子分布,茎、叶及分蘖芽鞘表面均有孢子存在。Cynodon dactylon Pers was a malignant weed in fields, gardens and lawn. A pathogenic fungus isolated from C. dactylon flower smut was identified and characterized to study its potential bioherbicidal application. Identification of the strain of LHL1 was carried out by using ITS rDNA sequence homology comparison as well as morphological traits. The carbon and nitrogen resources and initial pH value were optimized for its liquid culture. The distribution of spores and fungus in different parts of Cynodon dactylon were also studied. The results showed that strain LHL1 belonged to Basidiomycotina, Ustilaginales and Ustilago. Glycerol and Lactoalbumin hydrolysate were the optimal carbon and nitrogen resources for the fungus growth during the tests, and the optimal initial pH value was 5. A great amount of spores and fungus were found on different organs epidermis of host plant except root. Moreover, spores could be found on epidermis of leaf sheath, leaf, stem and germ sheath.
