摘 要:以野外采集的平菇为样本,经菌丝培养后,用FDEB法、FDEB-CTAB法、改良CTAB法和改良SDS法提取DNA,并进行琼脂糖凝胶电泳检测、紫外分光光度计分析和标准差及标准误分析,结果表明:FDEB法提取的平菇DNA产率最高,但存在较严重RNA的污染,标准差和标准误比较大,稳定性差;改良CTAB法综合效果不错,提取稳定性高;改良SDS法提取的平菇DNA的纯度最好,标准差和标准误最低,提取效果稳定性最好,但产量相对较低;FDEB-CTAB法提取平菇DNA,去多糖的效果很好,标准差和标准误适中,但产量较低。综合以上结果,改良CTAB法提取蘑菇DNA综合效果最好。By mycelium culture of wild Pleurotus ostreatus,its genomic DNA was extracted by four methods,which were the FDEB,FDEB- CTAB,modified CTAB and improved SDS,which was analyzed by agarose gel electrophoresis,ultraviolet spectrophotometer,and standard deviation and standard error of arithmetic mean. The results showed that the yield of DNA by FDEB was the highest among four methods. But there was severe RNA pollution,and the change of standard deviation and standard error appeared a few fluctuations. The DNA quality by the modified CTAB was obviously better and stable. The purity of DNA by the improved SDS was the highest and change of standard deviation and standard error was the lowest. Whereas its yield was relatively low. The polysaccharide pollution could be removed effectively by FDEB- CTAB method,whose change of standard deviation and standard error was moderate. But yield was very low. So the modified CTAB was the best method for extracting genomic DNA from Pleurotus ostreatus.
