摘 要:目的探讨噪声习服对豚鼠耳蜗内miRNA183表达的影响,及其在习服保护作用中可能的作用机制。方法 40只雄性豚鼠随机分为正常对照组(A组),噪声习服组(B组),噪声习服+强噪声暴露组(C组),单纯强噪声暴露组(D组),采用耳蜗基底膜铺片的方法测定各组毛细胞缺失率的变化,采用Real-time PCR方法检测各组豚鼠耳蜗中miRNA183的表达水平。结果耳蜗基底膜铺片显示,B组耳蜗毛细胞缺失率与A组差异无统计学意义,C组与D组相比基底膜毛细胞缺失显著减少(P<0.05);Real-time PCR结果显示,B组miRNA183相对表达量较A组升高(98.1±4.79)%(P<0.05),D组miRNA183表达量较A组下降达(77.4±3.97)%,D组miRNA183相对表达量较C组下降(71.4±1.57)%(P<0.05)。结论无损伤的中等强度习服声可诱导耳蜗中miRNA183的表达,并减少其后强噪声暴露引起的miRNA水平下调及毛细胞缺失,从而产生听力保护作用。习服过程所产生的听力保护可能与miRNA183水平升高有关。Objective To observe the expression characteristic of miRNA183 in cochlea of guinea pigs after low level sound conditioning.Methods Forty male healthy guinea pigs with normal Preyer's reflex were randomly divided into four groups:normal group(group A),conditioning noise group(group B),conditioning noise followed by high-level noise group(group C),high-level noise group(group D).Hair cell loss was measured by surface preparation technique.The expression of miRNA183 in guinea pigs cochleae after exposure to noise was examined by Real-time PCR.Results Cochlear surface preparations indicated that the loss of hair cells induced by noise exposure was obvious.A significant reduction of the loss of hair cells was observed between group D and group C.The expression of miRNA183 in group B showed a significant increase compared with that in group A(98.1±4.79 % reduction;P〈0.05),the expression of miRNA183 in group D showed a significant reduction compared with that in group A and C(77.4±3.97%and 71.4±1.57%reduction respectively;P〈0.05, Fig.1).Conclusions MiRNA183 in cochlea induced by sound conditioning is higher than that in normal cochlea organ,sound conditioning can decrease hair cell loss induced by the following high-level noise exposure,and miRNA183in cochlea organ induced by sound conditioning may provide protective effect on auditory system.
