In vitro Multiple Screening of Antagonistic Bacillus against Phoma sp. and Optimizing Its Fermentation Factors

摘  要:以赞皇大枣(Ziziphus jujuba‘Zanhuang Dazao’)黑腐病主要病原菌茎点霉菌(Phoma sp.)为靶标,对分离自土壤中的10株芽孢杆菌(Bacillus spp.)菌株进行室内多重筛选,并通过单因素试验对筛选获得的目标菌进行发酵培养基和发酵条件优化。结果表明,芽孢杆菌菌株B01对靶标菌有明显而稳定的拮抗效果;其最佳发酵培养基配方为蔗糖20.0 g/L,花生粉26.0 g/L,酵母粉5.5 g/L,KH2PO40.3 g/L,MgSO4·7H2O0.2 g/L,CaCO30.4 g/L,FeSO4·7H2O 0.02 g/L,MnSO4·7H2O 0.02 g/L;种子液浓度1.5×10^7~2.0×10^8CFU/mL时,最佳发酵条件为接种量4%(体积比),装液量250 mL三角瓶装20 mL,原始pH 6.0,发酵温度24℃,发酵时间48 h。Antagonistic Bacillus against Phoma sp. was selected by multiple screening in vitro. The optimal fermentation medium and conditions for the strain selected were determined with single-factor experiment design. The results showed that strain Bacillus B01 had obvious and stable antagonistic effect on Phoma sp. The optimal fermentation medium for strain B01 consists of sucrose, 20 g / L; peanut powder, 26.0g / L; yeast extract powder, 10g / L; KH2PO40.3g / L; MgSO4·7H2O 0.2 g / L; CaCO3 0.4 g / L; FeSO4·7H2O 0.02 g / L; MnSO4·7H2O 0.02 g / L; The optimal fermentation conditions were defined as follows: concentration of seed broth, 1.5×10^7~2.00×10^8CFU / mL; inoculation amount, 4%(V / V); initial pH at 6.0; the volume of broth in a 250 mL flask, 20 mL; fermentation temperature, 24 ℃; fermentation time, 48 h.
