中药清热利湿饮对HaCaT细胞周期的影响

摘　　要：目的探讨中药清热利湿饮水提液对经肿瘤坏死因子α(TNF-α)诱导活化的人永生化角质形成细胞(HaCaT)细胞周期的影响。方法采用MTT法检测TNF-α对HaCaT细胞增殖的影响,流式细胞仪技术检测清热利湿饮提取液对HaCaT细胞周期的影响。结果经2.5×105 u/L的TNF-α诱导的HaCaT细胞增殖率比对照组增加了(14.11±2.97)%,12.5 g/L和15 g/L清热利湿饮可使HaCaT细胞阻滞于S期,与TNF-α对照组相比,S期比率分别增加了10.63%和21.55%。结论 TNF-α可使HaCaT细胞活化及过度增殖,一定浓度的清热利湿饮以浓度依赖的方式干扰正常的HaCaT细胞周期,通过阻滞细胞的周期时相而抑制HaCaT细胞的分裂和过度增殖。Objective To investigate the influence of extractions that got from the Chineses herb Qingre Lishi Yin on cell cycle of keratinocyte HaCaT cells induced by TNF-α.Methods The influence of TNF-α on HaCaT cells proliferation was measured by MTT assay.The influence of Qingre Lishi Yin on HaCaT cell cycle and apoptosis were analyzed by flow cytometry.Results The HaCaT value of cells induced by 2.5× 10^5 u/L TNF-α was（14.11 ±2.97）% more than the control group,12.5 g/L and 15 g/L Qingre Lishi Yin could make HaCaT cell cycle arrest in S phase compared with control group TNF-α.S ratios were increased 10.63% and 21.55%.Conclusion TNF-α can make the HaCaT cell activate and proliferate,and a certain concentration of Qingre Lishi Yin interfere with normal cell cycle of HaCaT cells in a concentration-dependent mode,and inhibit cell division and proliferation through arrestting cell cycle phase of HaCaT.
