HPLC法同时测定细辛提取物及其配方颗粒中细辛脂素等5种成分的含量

摘　　要：目的建立细辛提取物及其配方颗粒中细辛脂素、芝麻脂素、甲基丁香酚、卡枯醇和2-甲氧基-4,5-亚甲二氧基苯丙酮共5种成分的含量测定方法。方法采用HPLC法。色谱柱为Inertsil ODS-3柱（250 mm×4.6 mm,5μm）,流动相为甲醇-水,梯度洗脱,流速为1.0 mL·min-1,检测波长为287 nm,柱温为35℃。结果细辛脂素、芝麻脂素、甲基丁香酚、卡枯醇和2-甲氧基-4,5-亚甲二氧基苯丙酮质量浓度分别在7.5~300 mg·L-1、4~160 mg·L-1、2~80 mg·L-1、0.9~90 mg·L-1和3~300 mg·L-1内与峰面积呈良好的线性关系（r〉0.999 3）;平均加样回收率（n=6）在97.5%~101.0%内,RSD均小于1.99%。不同细辛提取物及配方颗粒中5种成分含量差异较大。结论本方法准确、重复性好,可用于细辛提取物及其配方颗粒的质量控制。Objective To establish a method for the simultaneous determination of（-）-asarinin,（-）-sesamin, methyleugenol,kakuol and 2-methoxyl-4,5-methylene-dioxypropiophenone in the extracts and formula granules of Asari Radix et Rhizoma. Methods An HPLC method w as performed on an Inertsil ODS-3 column（ 250 mm × 4. 6 mm,5 μm） with the column temperature at 35 ℃. The mobile phase consisted of methanol-w ater in a gradient elution mode at the flow rate of 1. 0 mL·min- 1. The detection w avelength w as set at 287 nm. Results The linear ranges of（-）-asarinin,（-）-sesamin,methyleugenol,kakuol and 2-methoxyl-4,5-methylene-dioxypropiophenone show ed a good linearity in the range of 7. 5- 300 mg·L- 1,4-160 mg·L- 1,2-80 mg·L- 1,0. 9-90 mg·L- 1and 3-300 mg·L- 1,respectively. The mean recoveries of 5 marker components ranged from 97. 5% to 101. 0% w ith RSD less than 1. 99%. The results show ed that there w as a significant difference in the contents of the marker components betw een the commercial extracts and formula granules. Conclusions The method is accurate,reproducible and practical for the quality control of the extracts and formula granules of Asari Radix et Rhizoma.
