摘 要:目的:观察小青龙汤对变应性鼻炎(AR)大鼠血清IgE、IL-12、LTC4水平的影响。方法:通过卵白蛋白全身致敏及局部激发法建立过敏性鼻炎大鼠模型,另设空白对照组和实验用药高、中、低剂量组,通过比较各组间症状积分的差异确立最佳用药浓度,并采用ELISA法测定各组血清IgE、IL-12、LTC4水平。结果:模型组的症状积分与空白组相比具有显著性差异(P<0.01),各治疗组症状积分明显低于模型组(P<0.01),高中低剂量组之间比较,高剂量组优于中、低剂量组(P<0.05),确立为最佳用药剂量组。模型组血清中IgE、LTC4含量明显升高,IL-12含量明显降低,与空白组相比有显著性差异(P<0.01),小青龙组血清中IgE、LTC4水平明显降低,IL-12水平明显升高,与模型组相比有显著性差异(P<0.01).结论:小青龙汤对AR大鼠症状具有明显的改善作用,尤以高剂量组最为突出,其多靶点抗鼻过敏反应作用,可能是通过提高TH1类细胞因子IL-12含量,使TH1/TH2比恢复平衡,降低IgE水平,抑制过敏反应慢反应物质LTC4释放,实现降低炎性介质分泌从而改善鼻粘膜水肿,缓解鼻炎症状。Objective: To study the effect of Xiaoqinglong Decoction on serum IgE, IL-12, LTCA level in allergic rhinitis Rats. Methods: Male SD rats were randomly divided into normal group, model group, Xiaoqinglong high dose group, middle dose group and low dose group. Ovalhumin systemic sensitization and local attack in rats AR model preparation. Observing symptoms in different groups and calculating scores, then determining the best drug concentration. Determining the serum IgE, IL-12, LTC4 level by ELISA method. Results: Compared with the normal group, sysptom integral of the model group was high significantly ( P 〈 0. 01 ) , symptom scores of the treatment groups were significantly lower than the model group ( P 〈 0. 01 ) , high dose group was better than other dose groups ( P 〈 0. 05 ), so we established it as the best dosage group. Compared with the normol group, the levels of IgE, LTC4 in model group were significantly increased, but IL-12 level was decreased significantly. However the Xiaoqinglong Decoction could elevate the level of IL-12, decrease [gE, LTC4 levels evidently. Conclusion: Xiao- qinglong Decoction has obvious effect to improve symptoms in AR rats, especially in high dose group, it has multi-target anti- nasal allergic reaction, through increasing the TH1 type eytokines of IL-12 content, TH1/TH2 than to restore balance, reduce IgE levels, inhibit allergic reactions slow reactive substances LTC4 release and reduce the secretion of inflammatory mediators in order to improve nasal edema, alleviate the symptoms of rhinitis.
