摘 要:目的:观察"祛风通窍方"对血管性痴呆大鼠海马线粒体SOD、MDA变化的影响及作用机制。方法:用永久性结扎双侧颈总动脉法制备血管性痴呆大鼠模型,Morris水迷宫筛选符合要求SD大鼠50只,随机分为模型组、尼莫地平组、祛风通窍方高、中、低剂量组每组各10只,假手术组10只。造模1月后,尼莫地平组予尼莫地平6.25mg/kg/d灌胃,祛风通窍方高、中、低剂量组分别给予祛风通窍方26.8g/kg/d、13.4g/kg/d、6.7g/kg/d灌胃,假手术组、模型组予等体积蒸馏水灌胃,连续30天。Morris水迷宫检测其行为学后,断头处死取海马组织,梯度离心获得线粒体,化学比色法检测SOD、MDA变化。结果:行为学情况:随着训练时间延长,各治疗组大鼠逃避潜伏期(EL)缩短,平台象限停留时间延长,其中以阳性组,祛风通窍方高、中剂量组明显,差异有统计学意义(P<0.05)。SOD、MDA变化:与假手术组比较,模型组、阳性组及高、中、低剂量组SOD活性降低,MDA含量升高,差异有统计学意义(P<0.05);与模型组比较,阳性组及高、中剂量组SOD活性升高,MDA含量降低,差异有统计学意义(P<0.05);与阳性组比较,高、中剂量组SOD活性增强,MDA含量降低,差异有统计学意义(P<0.05);与低剂量组比较,高、中剂量组SOD活性升高、MDA含量降低,差异有统计学意义(P<0.05)。结论:祛风通窍方改善血管性痴呆大鼠的行为学;可增强SOD活性、减少MDA含量,发挥抗氧化作用,进而改善血管性痴呆大鼠的认知功能。Objective : To observe the impact and the mechanism of Qufeng Tongqiao Prescription ( expelling wind and promoting the restoration of consciousness) to the SOD, MDA changes of hippocampus mitochondrial of rats with vascular dementia. Methods: Vascular dementia rats were prepared through a permanent bilateral common carotid artery ligation. After Morris water maze screening, 50 SD rats that met the requirements were selected and randomly divided into model group, Nimodipine group, Qufeng Tongqiao high, medium, and low-dose groups, 10 rats each, and another 10 rats in the sham operation group. After one month, through garage the Nimodipine group was given Nimodipine 6. 25mg/kg/d, the Qufeng Tongqiao high, medium, and low-dose groups were given Qufeng Tongqiao prescription 26. 8g/kg/d, 13.4g/kg/d, and 6. 7g/kg/d, and the sham operation group and model group were both given an equal volume of distilled water for 30 days. After their behavior were checked by Morris water maze, the rats were decapitated and removed hippocampus, which went through gradient centrifugation to obtain mitochondria and then chemical colorimetric to test SOD, MDA changes. Results: Behavioral situation: with the extension of training time, the escape latency (EL) of the treated rats became shortened and the platform quadrant stayed longer, especially for the positive symptoms group, the Qufeng Tongqiao high and medium dose groups, whose differences were statistically significant (P 〈 0.05). SOD, MDA change: compared with the sham operation group, the model group, the positive symptoms group and the high, medium and low dose groups showed a decreased SOD activity while their MDA content increased, and the differences were statistically significant (P 〈 0. 05) ; compared with the model group, the positive symptoms group and the high and medium dose groups showed an improved SOD activity while their MDA content decreased, and the differences were statistically significant ( P 〈 0. 05 ) ; compared with the posit
