基因芯片技术在阴道分泌物常规检测中的应用研究

摘　　要：目的探讨基因芯片技术在阴道分泌物常规检测中的应用价值。方法选取2012年1—4月首都医科大学附属北京天坛医院妇产科门诊400例患者阴道分泌物,每份标本同时用BD Affirm VPⅢ芯片检测、显微镜镜检、细菌性阴道病(BV)甲酚蓝试剂检测和BV蓝试剂检测,并接种至念珠菌显色培养基和滴虫培养基培养。采用SPSS 16.0统计软件,行Kappa检验。结果以BV蓝试剂检测结果为金标准,BV甲酚蓝试剂检测和BD Affirm VPⅢ芯片检测BV的Kappa值分别为0.813和0.553。以念珠菌培养结果为金标准,显微镜镜检和BD Affirm VPⅢ芯片检测念珠菌的Kappa值分别为0.605和0.941。以滴虫培养结果为金标准,显微镜镜检和BD Affirm VPⅢ芯片检测滴虫的Kappa值分别为0.973和0.923。结论 BD Affirm VPⅢ芯片检测念珠菌和滴虫一致性较好,BD Affirm VPⅢ芯片与BV蓝或BV甲酚蓝试剂联合用于临床进行阴道分泌物检查,可以取代显微镜镜检。Objective To study the application of gene chip technique in leucorrhea routine examination.Methods Vaginal secretions of 400 gynecological outpatients referred to our hospital from January 2012 to April 2012 were tested with BD Affirm VPⅢ chips,microscopes,BV cresol blue agent and BV blue agent for testing bacterial vaginopathy respectively,which were cultured in candida color medium and trichomonad culture mediums.Kappa tests were performed with the statistical program SPSS 16.0.Results The results of BV blue reagent were regarded as golden standard for testing bacterial vaginopathy,the Kappa values of BV cresol blue reagent and BD Affirm VPⅢ chips were 0.813 and 0.553 respectively.The results of candida chromogenic plates were regarded as golden standard for testing candida,the Kappa values of microscope and BD Affirm VPⅢ chips were 0.605 and 0.941 respectively.The results of trichomonad culture mediums were regarded as golden standard for testing trichomonad,the Kappa values of microscope and BD Affirm VPⅢ chips were 0.973 and 0.923 respectively.Conclusion The BD Affirm VPⅢ gene chips accompanied with BV blue reagent or BV cresol blue reagent can take the place of microscope examination to test vaginal discharge clinically.
