摘 要:目的探讨GSTO1基因多态性与饮水型地方性砷中毒易感性的关系。方法对新疆奎屯砷中毒病区慢性砷中毒患者96人,病区内对照组73人,以及非病区外对照组89人,应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对GSTO1基因rs4925及rs11509438多态位点进行检测,用聚合酶链反应-双对引物多态性(PCR-CTPP)方法对GSTO1基因rs11509437位点进行检测,分析不同基因型与砷中毒发病风险的关系。结果 GSTO1基因rs4925、rs11509438及rs11509437位点的基因型以及等位基因频率在三组间的分布差异均无统计学意义(P>0.05),GSTO1基因rs4925、rs11509438及rs11509437位点的基因多态性在不同砷暴露人群中的发病风险,差异均无统计学意义(P>0.05)。结论 GSTO1基因rs4925、rs11509438及rs11509437位点可能与新疆饮水型地方性砷中毒无关联。Objective To investigate the relationship between genetic polymorphism in GSTO1 with water-born endemic arsenism. Methods GSTO1 gene polymorphism were analyzed by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) and Polymerase Chain Reaction- Confronting Two-pair Primer (PCR-CTPP). Analysis was carried out among 96 arsenism patients (the case group), 73 healthy individuals (the intra-control group) and 89 unrelated healthy individuals (the extra-control group). Results The distributions of genotype frequencies of GSTO1 gene SNPs (rs4925, rs11509437 and rs11509438) were no statistically significant (P〉0.05); GSTO1 gene SNPs (rs4925, rs11509437 and rs11509438) couldn't increase arsenism risk among the three groups (P〉0.05). Conclusion GSTO1 gene SNPs (rs4925, rs11509437 and rs11509438) may not be associated with water-horn endemic arsenism.
