摘 要:目的探讨白藜芦醇对脂多糖(LPS)致人脐静脉血管内皮(HUVEC)细胞凋亡保护效应以及对凋亡相关蛋白Bax影响。方法 LPS 1μg/ml刺激HUVEC细胞24 h建立凋亡模型,之后给予白藜芦醇160,80,40,20μg.L-1干预,CCK-8法检测细胞活力,Annexin-V/FITC双染后流式细胞仪检测细胞凋亡率,Western blot方法检测凋亡相关蛋白Bax表达。结果与模型组比较,白藜芦醇呈剂量依赖性的提高LPS诱导的HUVEC细胞存活率,降低HUVEC细胞凋亡率,降低Bax蛋白表达,有统计学差异(P<0.05)。结论白藜芦醇对LPS致HUVEC细胞凋亡有保护效应,其效应可能与抑制Bax表达有关。Objective To investigate the protect effects of resveratrol and the protein expression of Bax on apoptosis of HUVEC ceils induced by LPS. Methods Apoptosis of HUVEC cells was induced by LPS 1 μg/ml for 24 hours. 160, 80, 40, 20 μ g.L-1 resveratrol were used for intervention. Cell viability was observed by CCK-8. Apoptosis was tested by the flow cytometry. Protein expression of Bax was tested by Western blot. Results Compared with model group, resveratrol could increase the cell viability, decrease the apoptosis and Bax expression, which was dose depended. Conclusion Resveratrol could retard the apoptosis of HUVEC cell induced by LPS, and its mechanism may be associated with decreasing of the protein expression of Bax.
