摘 要:目的:研究甘氨双唑钠(CMNa)对人卵巢癌细胞株SKOV3的放射增敏效应及作用机制。方法:MTT法检测CMNa对SKOV3细胞增殖的影响,并筛选出药物的无毒浓度范围;以不同浓度CMNa+放射线联合作用于SKOV3细胞为实验组,单纯照射SKOV3细胞为对照组,应用MTT法检测其放射增敏效应,运用流式细胞仪分析细胞周期分布及活性氧(ROS)含量的变化。结果:SKOV3细胞的存活率随CMNa作用浓度的增大及作用时间的延长而下降,选择进行放射增敏实验的高、中、低质量浓度为0.05,0.20,0.80 mg·ml-1,其可显著增强SKOV3细胞的放射敏感性,且随浓度的增大,其增敏效应显著性增强(P<0.01),增敏比值分别为1.29,1.63和1.87;流式细胞仪检测结果显示,在低剂量(2 Gy)时,CMNa可将SKOV3细胞阻滞在G2/M期,浓度越大,G2/M期阻滞越严重(P<0.05),且随浓度增大,SKOV3细胞内的ROS含量增加越多(P<0.05)。结论:一定浓度范围的CMNa对SKOV3细胞具有显著的放射增敏效应,其增敏效应随药物浓度的增加而增强,其放射增敏机制可能与将SKOV3细胞阻滞在放射敏感性最高的G2/M期以及增加对细胞有氧化性损伤的ROS含量有关。OBJECTIVE To study the radiosensitive effect and mechanism of sodium glycididazole (CMNa) on human ovari- an cancer cell line SKOV3. METHODS MTT assay was used to investigate the effect of CMNa on proliferation of SKOV3 ceils and select its range of the non-toxic concentrations. The SKOV3 cells were divided into radiation group as control group, and CMNa of different concentrations combined with radiation as experimental group. The radiosensitive effect of CMNa was detec- ted by MTT assay. Changes of cell cycle and ROS content in SKOV3 cells after different treatments were analyzed by flow cy tometry. RESULTS The survival rate of SKOV3 cells decreased when the concentration and treatment time of CMNa in creased. The high,medium and low concentrations of CMNa in radiosensitization experiment were 0. 05,0. 20,0. 80 mg-ml which could significantly enhance the radiosensitivity of SKOV3 cells. The radiosensitive effect enhanced with the increase of concentration of CMNa (P〈0. 01 ). Their radiosensitization ratios were 1.29,1.63 and 1.87. Flow cytometry analysis showed SKOV3 cells treated with CMNa were accumulated in G2/M phase. The proportion of G2/M phase and ROS content in SKOV3 cells increased with the increase of concentration of CMNa when the radiation dose was 2Gy (P〈0. 05). CONCLUSION CM- Na has obvious radiosensitive effect on human ovarian cancer ceil line SKOV3, and the effect increases with the increase of con- centration. The mechanism of its radiosensitive effect may be associated with the accumulation of SKOV3 cells in G2/M phase which cells are radiosensitive and the increase of ROS content which has oxidative damage effect on SKOV3 cells.
