摘 要:目的观察黄芪对糖尿病大鼠血清胰高血糖素样肽-1(GLP-1)及炎症因子肿瘤坏死因子-α(TNF-α)、白介素-18(IL-18)的影响。方法选取雄性大鼠60只,随机选取40只以链脲菌素(STZ)30 mg/kg制成糖尿病大鼠模型。余下20只正常喂养,设为对照组。将成模糖尿病大鼠随机分为2组:模型对照组与黄芪治疗组。将60 mg/mL黄芪药液按200 mg/kg体质量给药,同时模型对照组和对照组灌胃生理盐水。2周后给予3组大鼠葡萄糖液灌胃,30 min后尾静脉采血。应用酶免法(ELISA)方法测定两组大鼠空腹血清GLP-1、IL-18、TNF-α水平及大鼠服糖后30 min血清GLP-1水平。结果模型组及黄芪组血清空腹及糖负荷后GLP-1水平低于对照组、TNF-α、IL-18水平高于对照组(P<0.01)。黄芪组血清空腹及餐后GLP-1水平高于模型组、TNF-α、IL-18水平低于模型组(P<0.05)。GLP-1水平与IL-18、TNF-α呈负相关(相关系数r=-0.64,P<0.05;r=-0.37,P<0.05)。结论黄芪可升高2型糖尿病大鼠血清GLP-1水平,GLP-1水平的升高可能与IL-18、TNF-α相关。Objective: To investigate the effects of astragalus membranaceus on GLP-1 ,TNF-α,and IL-1 of diabetic rats serum. Methods: 60 rats were selected, among which 40 were taken to establish model of diabetic rats by injecting streptozotocin (STZ) 30 mg/kg. The rest 20 rats were given normal feeding as control group. The model of diabetic rats was randomly divided into 2 groups: model control group and astragalus membranaceus treatment group. The 60 mg/mL solution of astragalus membranaceus was given 200 mg/kg administration. At the same time ,the model control group and the control group were fed with 0.9% saline. Three groups of rats received intragastric administration of glucose. 30 minutes later, blood was taken through tail vein. GLP-1 in serum before and 30 min after sugar taken,IL-18,and TNF-α levels of rats were measured by ELISA. Results: GLP-1 in serum level before and after glucose load in model group and astragalus membranaceus group were lower than that in control group. TNF-α and IL-18 levels were higher than those in control group (P〈0.01). GLP-1 in serum level before and after glucose load in astragalus membranaceus group was higher than that in model group. TNF- α and IL-18 levels of astragalus membranaceus group were lower than those in model group (P〈 0.05). GLP-1 was negatively correlated with IL-18 and TNF-α (correlation coefficient r=-0.64,P〈0.05 ,r=-0.37,P〈0.05. The difference has statistical significance). Conclusion: Astragalus membranaceus can elevate serum GLP-1 levels in serum of rats with type 2 diabetes. Elevated level of GLP-1 may relate to IL-18 and TNF-α.
