地龙、乌梢蛇对系膜增生性肾炎模型大鼠肾组织TLR4、TGF-β_1表达的影响

摘　　要：目的观察地龙、乌梢蛇对系膜增生性肾炎(MsPGN)大鼠的治疗作用及对Toll样受体4(TLR4)、转化生长因子β1(TGF-β1)表达的影响。方法选用SPF级Wistar大鼠60只,采用改良慢性血清病法复制MsPGN模型并分组,各治疗组大鼠分别予不同剂量地龙、乌梢蛇颗粒制剂灌胃,正常对照组、模型组予等量生理盐水灌胃。治疗8周,检测各组大鼠24 h尿蛋白定量,HE染色观察各组大鼠肾组织病理改变;免疫组化(SABC)法检测TLR4、TGF-β1在各组大鼠肾组织的表达。结果模型对照组24 h尿蛋白定量明显高于正常对照组(P<0.01),地龙、乌梢蛇各治疗组尿蛋白定量明显低于模型对照组(P<0.05),TLR4、TGF-β1的表达模型组较正常组增多(P<0.01),各治疗组较模型对照组明显减少(P<0.01)。结论地龙、乌梢蛇能减少(MsPGN)模型大鼠蛋白尿,改善肾脏病理变化,这可能与抑制肾脏组织中TLR4、TGF-β1的过度表达有关。Objective： To observe the effects of Lumbrici and Zaocys dhumnades on rats with mesangial proliferative glomerulonephritis （MsPGN） and on expressions of Toll receptor 4 （TLR4） and transforming growth factor （TGF-β）. Methods： 60 Wistar rats of clean grade according to MsPGN model were established by method of chronic serum disease improvement,and randomly divided into normal control group,model group,and groups of Lumbrici and Zaocys dhumnades of different doses. The normal group and model group were given by gavage with equal volume of water while other groups were given by garage with different doses of Lumbrici and Zaocys dhumnades. Rats 24 hours urine protein and pathological changes of renal tissue were observed in each group by HE staining 8 weeks later. Toll receptor 4 and transforming growth factor in renal tissue were also observed by immunohistochemistry. Results： The urine protein in 24 hours was higher in model control group than that in normal control group（P〈0.01 ）. The quantity of urinary protein in treatment group was significantly lower than that in model group （P〈0.05）. Expressions of TLR4 and TGF-β1 in model group increased compared with normal group （P〈0.01）. Expressions of TLR4 and TGF-β1 in treatment group decreased significantly than those in control group （P〈0.01 ）. Conclusion： Lumbrici and Zaocys dhumnades can reduce proteinuria,and contribute to inhibiting expressions of TLR4 ,and TGF-βl in renal tissue.
