摘　　要：目的观察痰瘀同治代表药物丹蒌片药物血清对血小板衍生因子BB型（PDGF—BB）诱导下体外培养的血管平滑肌（VSMC）增殖的影响，并探讨其可能的机制。方法SD大鼠喂饲丹蒌片药物溶液收集药物血清。组织块贴壁法培养SD大鼠原代胸主动脉平滑肌细胞，取第3代细胞进行免疫荧光法鉴定，取4～7代细胞进行实验。MTT比色法观察正常血清培养及丹蒌片药物血清（低、中、高浓度）对PDGF—BB（25μg／L）所致的VSMC增殖的影响。结果MTT法显示与对照组相比，药物血清组对PDGF—BB诱导的VSMC增殖呈剂量依赖性抑制，高浓度与低浓度比较有明显差异（P＜0．05）。结论丹蒌片可有效抑制PDGF—BB诱导的VSMC增殖，提示其控制AS的过程可能与抑制平滑肌增殖并向内皮下迁移有关。Objective： To observe the effect of pharmacological serum of Danlou Pill,representive of method of combined phlegm and blood stasis treatment,on vessel smooth muscle cell （VSMC） proliferation in vitro induced with platelet derived growth factor-BB （PDGF-BB）,and to investigate the capable mechanism. Methods： SD rats were fed with Danlou Pill and pharmacological serum was extracted. Primary aortic vessel smooth cell of SD rats were cultured with the tissue explants adherent method ,the third generation cells were taken to identify, and the forth to seventh cell were taken to experimentize. The effect of normal serum and different concentrations （low, media and high） of pharmacological serum of Danlou Pill on VSMC proliferation induced by PDGF-BB（25μg/L） were observed with MTY colorimetry. Results： MTT colorimetry showed that VSMC proliferation was inhibited by way of dose-depend contrasted with control group. There were significant statistical difference between the high and low dose groups （P〈0.05）. Conclusion： Danlou Pill can effectively inhibit the proliferation of VSMC induced by PDGF-BB which indicates that the control of AS process of Danlou Pill may relate to its effect of inhibiting proliferation of VSMC and immigration to endothelium.
