摘 要:目的观察淫羊藿苷对大鼠脑梗死后神经修复的作用及相关机制。方法制备MCAO大鼠18只(模型对照组6只,淫羊藿苷低剂量组6只,淫羊藿苷高剂量组6只),另设空白对照组6只,每组随机选3只进行神经功能缺损评分,以神经功能改善最佳组定为有效组;对有效组利用real-time PCR方法检测基质细胞衍生因子-1(SDF-1)、CXCR-4基因量的变化。结果各组在2 h、1 d、7 d神经功能评分相近,低剂量组在14 d时存在差异,确定低剂量组为有效剂量组;RT-PCR提示14 d SFD-1、CXCR-4基因量显著提高,差异具有统计学意义(均P<0.05)。结论淫羊藿苷可以促进大鼠脑缺血后的神经功能恢复,其机制可能与淫羊藿苷上调SDF-1,CXCR4基因有关。Objective: To investigate effect and mechanism of icariin on nerve restoration after rat with cerebral infarction. Methods: 18 MCAO rats(6 in model control groups,6 in low dose groups,and 6 in high dose groups), and 6 rats in blank control group were established. 3 of each group were randomly selected for NIHSS and the best improvement group of neurological function was chosen as the effective group. Then expression level of genes SDF - 1 and CXCR-4 were detected by RT-PCR. Results: The neurological score did not have difference in 2 h, 1 clay,and 7 days in each group. Low dose group showed difference in 14th day suggesting itself to be the effective dose group. RT-PCR indicated the expression level of genes SDF - 1 and CXCR-4 increased significantly in 14th day and the difference was statistically significant (all P〈0.05). Conclusion: Icariin can improve neurological function recovery after rat with cerebral ischemia, most likely via up-regulations of genes SDF-1 and CXCR-4.
