摘 要:目的研制并验证战地ABO血型检测的新方法。方法将3 cm×9 cm长的滤纸条对折成两半,于左右两侧下1/3中点处分别用红笔标上字母"A"和"B";距字母下缘约1 cm处,先滴1滴与字母相对应的单克隆抗体溶液(抗A、抗B凝集素),然后各加1滴抗凝静脉血于两侧抗体中央;将滤纸垂直立于盛有生理盐水的烧杯内,每隔1 min记录滤纸的变化,随后用玻片法检测该标本的血型;最后用以上方法检测100例未知血型的抗凝静脉血,验证滤纸检测结果是否跟玻片法完全一致。结果未发生凝集反应的一侧(非凝集侧)出现明显的红色条带,红色字母被遮住;发生凝集反应的一侧(凝集侧)未见明显的红色条带,字母未被遮住。随着时间的增加,非凝集侧条带长度逐渐增加至最大值;凝集侧条带长度基本维持在"0"水平线。第3、6、9 min时,非凝集侧的条带长度[(1.50±0.31)cm,(2.58±0.36)cm,(2.99±0.48)cm]与凝集侧的长度[(0.01±0.03)cm,(0.02±0.06)cm,(0.02±0.06)cm]比较差异均具统计学意义(P<0.01)。凝集侧各时间段的条带长度差异无统计学意义(P=0.55),非凝集侧各时间段的条带长度差异具统计学意义(P<0.01)。红细胞抗原与单克隆抗A或抗B抗体反应后,玻片上有凝集现象的94处反应,滤纸上均无红色条带;玻片上无凝集现象的106处反应,滤纸上都出现明显的红色条带,用滤纸检测的结果(A型31例,B型35例,AB型14例,O型20例)与玻片法完全一致。结论本方法以滤纸为反应介质,结果形象具体、真实可靠,为条件受限的战争环境下的ABO血型检测提供了一种新方法。Objective To invent and verify a new ABO grouping method for battlefield environment. Methods Filter paper (3 ~ 9 em2) was prepared before writing red letter "A" and "B" in the middles of left and right margins; a drop of cmxesponding anti- body was respectively spotted at the site which was 1 cm lower than the letter and then a drop of blood was spotted in the center of antibody. After that we put the paper into a beaker which contained NaC1 solution. The outcomes were recorded every minute. To verify our new method, slide method was used to test 100 blood samples as well. Results Red strap was observed in non-agglutinated side while no red strap was seen in agglutinated side. Red letter in non-agglutinated side was covered while agglutinated side was not. With time went on, red strap in non-agglutinated side became longer. At minute 3, 6 and 9, strap length in non-agglutinated side ( 1.50 ± 0.31, 2.58 ± 0.36, 2.99 ± 0.48 cm) was statistically different ( P 〈 0.01 ) from that in agglutinated side (0.01 ±0.03, 0.02 ± 0.06, 0.02 ± 0.06 cm). None of the the 94 agglutinated cases in slide were showed strap in filter paper, while all the 106 non-agglutinated cases in slide had obvious strap in paper. The results from filter paper method were totally consistent with that from slide method. Conclusion Conclusion The results from the new method are reliable, which offers a new idea for blood grouping in battlefield environment.
