摘  要:目的对国产及进口MEM培养基培养MRC-5细胞的效果进行比较。方法用国产及进口MEM培养基培养37代MRC-5细胞,于显微镜下观察培养24、48、72 h后的细胞的生长情况,并对细胞在33~37代传代过程中的细胞活性进行检测;以0.008 MOI的Oka株水痘-带状疱疹病毒感染37代MRC-5细胞,观察细胞病变情况。结果国产及进口MEM培养基培养的37代MRC-5细胞,细胞形态、贴壁、伸展、牵手、成网、单层等生长情况均相似,细胞数量也无明显差异,均可使细胞保持较高的活力,两组间细胞存活率比较,差异无统计学意义(P〉0.05);细胞病变均比较典型、广泛,病变程度均在70%以上,且细胞完整,未见明显撕裂现象。结论国产及进口MEM培养基均可满足MRC-5细胞的生长和病毒培养的宿主条件。Objective To compare the efficacies of culture of MRC-5 cells by domestic and imported MEM. Methods The MRC-5 cells of passage 37 were cultured in domestic and imported MEM respectively. The cells after culture for 24, 48 and 72 h were observed for growth,of which those of passages 33 ~ 37 were determined for activity. MRC-5 cells of passage 37 were infected with Oka strain of varicella-zoster virus at MOIs of 0. 008,and observed for CPE. Results The MRC-5 cells cultured in domestic and imported MEM showed no significant difference in morphology, adherence, spreading,linkage,networking,monolayer formation,count and survival rate(P〉0. 05). All the cells showed high activity as well as typical and extensive CPE(more than 70%). The cells were intact,in which no obvious breakages of network were observed. Conclusion Both domestic and imported MEM were suitable for growth of MRC-5 cells and virus culture.
