早期急性心肌梗死患者全血中microRNA-1的表达

摘　　要：目的:探讨早期急性心肌梗死(AMI)患者全血中microRNA-1(miR-1)的表达情况。方法:选取AMI患者28例和同期住院患者26例(对照组)。在心肌梗死发作后的不同时间段:AMI患者入院时(T0)、8 h(T8)、12 h(T12)、24 h(T24)、7 d、14 d,采血液样本并应用实时定量PCR法对全血中miR-1的表达水平进行检测。应用ELISA法检测AMI不同时间血浆心肌肌钙蛋白的水平。结果:AMI患者全血中miR-1的表达水平在T0、T8、T12、T24、7 d时间段均较对照组明显升高(P<0.01);而AMI后第14天时其表达水平与对照组差异无统计学意义(P>0.05)。受试者工作特征曲线结果提示miR-1的ROC曲线下面积在T0、T8、T12、T24、7 d时受试者工作特征曲线下面积分别为0.839、0.831、0.828、0.835、0.774(P<0.01)。结论:全血中miR-1可能作为早期诊断AMI的生物标志物。Objective： To examine the whole blood microRNA-1（ miR-1） expression in early acute myocardial infarction（ AMI）patients. Methods： Twenty-eight patients with AMI were selected,and 26 patients in control group. In different time periods： after the onset of symptoms of myocardial infarction in patients with AMI admission,8 hours,12 hours,24 hours,7 days,14 days（ T0,T8,T12,T24,7 d,14 d）,the blood samples were collected and the expression level of miR-1 in whole blood was measured by real-time quantitative PCR,the plasma cardiac troponin I expression in AMI different time was detected by enzyme-linked immunosorbent assay.Results： The miR-1 expression level in whole blood of the patients with AMI at T0,T8,T12,T24,7 d period was significantly higher than in control group（ P〈0. 01）; while there was no statistical difference between the expression level of miR-1 after AMI 14 days between AMI patients and the patients in control group（ P 〉0. 05）. The curve of the receiver operating characteristic（ ROC） curve analysis results suggested that the corresponding area under ROC was 0. 839,0. 831,0. 828,0. 835,and 0. 774 when miR-1 under the ROC curve was at T0,T8,T12,T24and 7 d（ P 〈0. 05）. Conclusions： miR-1 in whole blood might be as a kind of biomarkers of early diagnosis in AMI.
