高效液相色谱法测定四环素片含量的方法改进

摘　　要：目的：建立高效液相色谱（HPLC）法测定四环素片的含量。方法采用Kromasil C18（250 mm ×4．6 mm ，5μm）色谱柱，以0．1 mol · L -1磷酸氢二铵-乙腈（84∶16）为流动相，柱温为35℃，流速为1．0 mL · min-1，检测波长为280 nm。结果四环素质量浓度在0．0240～0．7697 mg · mL -1范围内呈良好的线性关系，r＝1．000（n＝6），平均回收率为100．4％，RSD为0.8％（n＝3）。结论该方法操作简便、准确、重复性好，可用于四环素片的含量测定。Objective To establish an improved HPLC method for the determination of Tetracycline Tablets .Methods The separa-tion was performed on a Kromasil C18 （250 mm × 4 .6 mm ,5 μm） column ,with the mobile phase of 0 .1 mol · L -1 diammonium hydrogen phosphate-acetonitriles（84∶16） at a flow rate of 1 .0 mL · min-1 .The column temperature was set at 35 ℃ ,and the detection wavelength was 280 nm .Results Tetracycline presented a good linear correlation in the range of 0 .024 0-0 .769 7 mg · mL -1 （r=1 .000） ,the limit of detection was 21 ng · mL -1 ,and the average recovery was 100 .4% ,with RSD of 0 .8% （n=9） .Conclusion This method was simple ,accurate ,reproducible ,and suitable for the quality control of Tetracycline Tablets .
