摘 要:目的:探讨重组人源性白介素-2(recombinant human IL-2,rhIL-2)对耐多药结核(multidrug-resistant TB, MDR-TB)病人外周血CD4^+CD25^+Foxp3^+T细胞及转化生长因子-β(transforming growth factor-β,TGF-β)含量的影响。方法选择MDR-TB病人40例,20例化疗,20例化疗+rhIL-2皮下注射,流式细胞仪检测病人外周血CD4^+CD25^+Foxp3^+T细胞含量,ELISA试剂盒检测外周血TGF-β的表达。结果 MDR-TB病人外周血CD4^+CD25^+Foxp3^+T细胞与TGF-β含量显著相关,治疗后化疗+rhIL-2组外周血CD4^+CD25^+Foxp3^+T细胞及TGF-β含量分别下降(1.55±0.29)%和(18.97±2.11)%,与化疗组的(0.83±0.13)%和(10.98±1.04)%相比差异有统计学意义(P<0.05)。结论 CD4^+CD25^+Foxp3^+T细胞及TGF-β在MDR-TB的发病中起重要作用,应用rhIL-2能够降低外周血CD4^+CD25^+Foxp3^+T细胞及TGF-β的含量。Objective To investigate the effect of recombinant human IL-2 on the quantity of the peripheral blood CD4^+CD25^+Foxp3^+T cells and TGF-β in multidrug-resistant tuberculosis patients. Methods We selected multidrug-resistant TB patients, Flow cytometry was used to determine the quantity of CD4^+CD25^+Foxp3^+T cells in the peripheral blood. The level of TGF-β was detected by ELISA. Results The TGF-β level and the CD4^+CD25^+Foxp3^+T cells/CD4+cells ratio in the peripheral blood of patients with MDR-TB was signiifcantly correlated (r=0.80, P〈0.01). The quantity of CD4^+CD25^+Foxp3^+T cells and the level of TGF-βin the peripheral blood of patients of chemotherapy plus recombinant human IL-2 group signiifcantly decreased after the treatment when compared with that of chemotherapy group. Conclusion CD4^+CD25^+Foxp3^+T cell and TGF-βplay an im-portant role in the pathogenesis of multidrug-resistant TB. Recombinant human IL-2 has an effect on the quantity of the peripheral blood CD4^+CD25^+Foxp3^+T cells and TGF-βin multidrug-resistant tuberculosis patients.
