摘 要:目的探讨骨骼肌成肌干细胞C2C12在体外分化为肌管的最佳生长条件。方法把C2C12细胞接种于基质胶(matrigel)、Ⅰ型胶原(collagenⅠ)、多聚赖氨酸(PLL)预包被的24孔板,观察细胞生长情况并刺激分化5天后进行免疫荧光染色,分别计算肌球蛋白(myosin)阳性肌管的融合率。结果基质胶组和多聚赖氨酸组肌管融合率明显高于Ⅰ型胶原组(P<0.01),但是多聚赖氨酸组肌管相对较小且多核肌管数(≥3个核)相对较少。结论在3种基质中,基质胶是促进C2C12细胞分化融合的最佳基质,对于C2C12成肌提供了可靠保证。Objective To investigate optimal growth condition for the diferentiation of C2C12 ceils to myotubes on three different growth substrate in vitro. Methods The myoblast cells were resuspended in 24 - well culture plates which was pre - coated with matri- gel, and poly -lysine. The growth and differentiation of C2C12 were observed, then the cells were examined by myotubes marker Myosin with immunocytofluorescence 5 clays later,and myotubes fusion rate was calculated, respectively. Results The myotubes fusion rates of C2C12 cells in matrigel group and polylysine group was significantly higher than group collagen type - I (P 〈 0.01 ). However, the myotubes and multinucleated myotubcs number ( ≥ 3 nuclei) was relatively small in polylysine group. Conclusion In three kinds of matrix, Matrigel is the best growth substrate to promote the differentiation and fusion of C2C12 cell and guarantee a reliable myogenesis.
