摘  要:目的:探讨药物代谢酶(CYP2A6*1、*2、*3、*4)4型基因多态性与烟草代谢物的相关性及其与冠心病的关系。方法:随机选取湖南地区汉族经冠状动脉造影确诊的冠心病患者205例,包括吸烟者129例,不吸烟者76例;正常对照208例,包括吸烟者78例,不吸烟者130例。应用限制性片段长度多态性聚合酶链反应(PCR-RFLP)对4组对象的CYP2A6基因型分布频率进行分析。结果:正常对照组与冠心病组CYP2A6基因型的分布频率无显著性差异;吸烟组与不吸烟组CYP2A6*2和CYP2A6*3的分布频率无显著性差异;吸烟组中CYP2A6*1的分布频率大于不吸烟组;不吸烟组中CYP2A6*4分布频率大于吸烟组;吸烟组冠心病发病率大于不吸烟组。结论:吸烟是冠心病的危险因素。吸烟者和不吸烟者的冠心病发病率有显著差异。CYP2A6基因多态性与烟草代谢物相关的冠心病可能存在相关性。Objective:To explore the association among four polymorphisms of CYP 2A6(1,2,3and 4),tobacco metabolites and coronary heart disease.Method:There were 205patients with coronary heart disease including 129 smokers and 76non-smokers,and 208healthy adults including 78smokers and 130non-smokes selected randomly in Hunan Han population.Polymerase chain reaction-restriction fragment length polymorphism PCR-RFLP was used to genotype the CYP2A6gene polymorphisms.Result:The distributions of four genotypes between the coronary heart disease and healthy adults groups had no significantly difference.There was no significantly difference of CYP2A6*2and CYP2A6*3between smokers group and nonsmokers group.CYP2A6*1frequency was higher in smokers group than that in nonsmokers group.CYP2A6*4frequency was higher in nonsmokers group than that in smokers group.Morbidity of coronary heart disease was higher in smokers group than that in nonsmokers group.Conclusion:Smoking is a risk factor of coronary artery disease.The incidence of coronary heart disease has significant differences between smokers and nonsmokers.Between CYP2A6gene polymorphisms and tobacco metabolites may be related to coronary heart disease.
