摘 要:目的研究氯沙坦对异丙基肾上腺素诱发小鼠心室重构的干预作用。方法 60只小鼠随机均分为生理盐水对照(A)组、异丙基肾上腺素模型对照(B)组、氯沙坦5mg/kg(C)组、氯沙坦10mg/kg(D)组、氯沙坦20mg/kg(E)组和卡托普利阳性对照(F)组。14d后称重法测定左心室质量/体质量比(LVW/BW)、右心室质量/体质量比(RVW/BW)及全心质量/体质量比(HW/BW),放射免疫法检测小鼠心肌血管紧张素Ⅱ(AngⅡ)、血浆环磷酸腺苷(cAMP)水平,分光光度法检测小鼠心肌羟脯氨酸(Hyp)含量,HE染色法观察心肌组织形态学变化。结果与A组相比,B组LVW/BW、HW/BW以及AngⅡ、Hyp、cAMP水平增加(P<0.01),心肌细胞变性、坏死;而C、D、E组能明显改善B组各观察指标的变化(P<0.01或P<0.05)。结论氯沙坦能有效干预异丙基肾上腺素诱发小鼠的心室重构。Objective To investigate the effect of losartan on isoproterenol-indueed ventrieular remodeling in mice. Methods Sixty mice were equally randomized into six groups of A(treated with normal saline), B(isoproterenol model), C(treated with losartan 5 mg/kg), D(treated with losartan 10 mg/kg) ,E(treated with losartan 20 mg/kg) and F(treated with eaptopril 50 mg/kg). Fourteen days later, the ratios of left ventricle weight to body weight(LVW/BW), right ventricle weight to body weight(RVW/BW) and heart weight to body weight (HW/BW) were measured. The levels of angiotensin Ⅱ (Ang Ⅱ ) and cyclic adenosine monophosphate (cAMP) were detected by radioirnmunoassay,and the content of hydroxyproline(Hyp) was determined by spectrophotometry. Moreover,the morphological changes of myocardial tissues were observed by HE staining. Results Compared with group A, the values of LVW/BW and HW/BW and the levels of Ang Ⅱ, Hyp and cAMP were increased(P〈0. 01), and myocardial cells showed the degeneration and necrosis in group B,which were all significantly improved in groups of C, D and E(P〈0. 01 or P〈0. 05). Conclusion Losartan can effectively intervene isoproterenol-induced ventricular remodeling in mice.
