T淋巴细胞亚群、NK细胞及Thl/Th2细胞因子与先兆流产发病的关系

摘　　要：目的探讨T淋巴细胞亚群、自然杀伤(NK)细胞及辅助T细胞(Th)1/Th2细胞因子与先兆流产发病的关系。方法采用免疫荧光标记和流式细胞术检测40例不明原因先兆流产患者(A组)和25例正常妊娠妇女(B组)外周静脉血CD3+T、CD4+T、CD8+T细胞及NK细胞,采用ELISA法检测血清中Th1型细胞因子(TNF-α和IL-2)及Th2型细胞因子(IL-4和IL-10)的表达水平。结果两组CD3+T、CD4+T、CD8+T细胞百分比及CD4+/CD8+比较无统计学差异(P>0.05)。A组NK细胞百分比低于B组[(8.7±3.2)%vs.(19.6±8.1)%](P<0.05)。A组血清Th1型细胞因子TNF-α和IL-2表达水平高于B组(P<0.05)。结论细胞免疫异常及Th1/Th2型细胞因子的病理性失衡与先兆流产的发生有关联。Objective To explore the relationship between T lymphocyte subsets,natural killer （NK） cells,Th1/Th2 cytokines and threatened abortion. Methods CD3^＋ T,CD4^＋ T,CD8^＋T cells and NK cells in peripheral blood from 40 patients with unexplained threatened abortion（group A） and 25 cases of normal pregnant women（group B） were detected by immunofluorescence labeling and flow cytometry. Thl cytokines of TNF-α, IL-2, IL-4 and IL-10 were detected by ELISA. Results There were no significant differences in the percentages of CD3^＋ T, CD4^＋ T, CD8^＋ T cells and CD4^＋/CD8^＋ between two groups（P〉0. 05）. NK cells percentage in group A was lower than that in group B [（8.7 ± 3.2）% vs. （19.6±8. 1）%]（P〈0. 05）. TNFa and IL-2 in group A were higher than those in group B（P〈0. 05）. Conclusion Cell immune abnormalities and pathological imbalance of Th1/Th2 may be related with threatened abortion.
