摘 要:目的探讨Nogo-A及其受体(NgR)在大鼠颅脑损伤后损伤脑组织的表达变化。方法将136只SD大鼠按随机数字表法分成对照组、假损伤组、轻型颅脑损伤组、中型颅脑损伤组、重型颅脑损伤组,采用自制改良的Feeney's自由落体装置制作大鼠颅脑损伤模型;伤后12 h、24 h、3 d、7 d和14 d采用免疫组化染色法检测损伤脑组织中Nogo-A和NgR蛋白的表达。结果伤后各时间点,各型颅脑损伤组Nogo-A表达水平均明显高于假损伤组(P<0.05);重型、中型、轻型颅脑损伤组之间Nogo-A表达均有显著差异(P<0.05)。而NgR的表达水平则在伤后3、7、14 d各型颅脑损伤组才明显高于假损伤组(P<0.05),各损伤组之间也有统计学差异(P<0.05)。结论 Nogo-A和NgR在不同程度的损伤脑组织中表达差异显著,且有各自的表达规律。Objective To explore the expression of Nogo-A and NgR protein in traumatic brain tissues. Methods One hundred thirty-six SD rats were randomly divided into five groups, i.e. control group (n=8), sham traumatic brain injury (TBI) group (n=8), mild TBI group (n=40), mediate TBI group (n=40), severe TBI group (n=40). TBI models were made by Feeney free-fall device. The expression of Nogo-A and NgR protein in the injured brain tissue was determined by immunohistochemistry method. Results The levels of Nogo-A expressions were significantly higher in the severe TBI group than those in the mediate TBI group (P〈0.05), which significantly higher than those in the mild TBI group (P〈0.05) 12 hours and 1, 3, 7 and 14 days after TBI, which were significantly higher than those in the control and sham TBI groups (P〈0.05). The levels of NgR expressions were significantly higher in the severe TBI group than those in the mediate TBI group (P〈0.05), which significantly higher than those in the mild TBI group (P〈0.05), 3, 7 and 14 days after TBI, which were significantly higher than those in the control and sham TBI groups (P〈0.05). The levels of Nogo-A and NgR protein expressions reached the peak 7 days after TBI. Conclusions The expressions of levels of Nogo-A and NgR in the injured brain tissues were positively related to the severity of TBI.
