摘 要:目的 氧化苦参碱(Oxymatrine)具有抗氧化、抗炎、抗病原微生物、抗过敏、抗心律失常、抗肿瘤等药理活性.文中探讨氧化苦参碱在体内和体外抗血管生成的作用. 方法 实验分3组,氧化苦参碱溶于无菌蒸馏水中设氧化苦参碱组,分5个浓度梯度,分别为0.1、0.2、0.3、0.4、0.5mg/mL;0.2 mg/mL沙利度胺为阳性对照组,另设空白对照组.以不同浓度氧化苦参碱处理人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC),分别采用MTT法、Transwell小室迁移实验观察氧化苦参碱对HUVEC细胞增殖和迁移的影响;采用鸡胚绒毛尿囊膜(chick ehorio-allantoic membrane,CAM)模型,观察氧化苦参碱在体内的抗血管生成作用. 结果 0.1 mg/mL、0.2 mg/mL氧化苦参碱作用时小血管生成条数分别为(78.31±11.63)和(54.85±9.76)条,较空白对照组减少(P<0.05);当浓度为0.3 mg/mL以上时,氧化苦参碱作用时大血管条数分别为(3.28 ±0.72)、(2.95±0.69)和(2.26±0.47)条,小血管生成的条数为(51.25 ±9.13)、(46.33±8.28)和(42.78 ±9.25)条,较空白对照组减少(P<0.05).0.1 ~0.5 mg/mL浓度的氧化苦参碱较空白对照组对HUVEC细胞增殖和迁移均具有明显抑制效应,抑制率分别为23.2%、32.2%、46.7%、49.4%和50.6%,细胞迁移率分别为99.7%、82.3%、78.4%、65.3%和58.6%,差异有统计学意义(P<0.05). 结论 氧化苦参碱在体内、体外均具有抗血管生成作用.Objective Oxymatrine embodies of is an alkaloid extracted from the roots of sophora legume.The aim of the study was to investigate the antiangiogenic effect of Oxymatrine in vivo and vitro.Methods There were three groups in the experiment:oxymatrine group Human Umbilical Vein Endothelial Cells (HUVEC) were treated by different concentrations of oxymatrine (0.1,0.2,0.3,0.4,0.5 mg/mL),and the cell proliferation and migration rate were determined by MTT and transwell migration assay.Chick embryo chorioallantoic membrane (CAM) model was used to detect the inhibitory effect of oxymatrine on angiogenesis in vivo.Results When the concentration was 0.1 to 0.2 mg/mL,oxymatrine significantly inhibited the formation of small blood vessels ([78.31 ± 11.63]) vs ([54.85 ± 9.76]) (P 〈 0.05),and when the concentration was more than 0.3 mg/mL,all blood vessels were inhibited (P 〈 0.05).Moreover,oxymatrine with the concentration from 0.1 to 0.5 mg/mL significantly inhibited the proliferation and migration of HUVEC (P 〈 0.05).Conclusion Oxymatrine has obvious antiangiogenic effect both in vivo and vitro.
