摘 要:目的大鼠痛风性关节炎模型(acute gouty arthritis,AGA)在实际研究中应用较多,但不同品系大鼠对尿酸钠晶体的反应性不同,会对成模效果产生一定的影响。文中对比Wistar大鼠和SD大鼠对尿酸钠结晶导致的AGA的敏感程度。方法将Wistar大鼠(32只)和SD大鼠(32只)按随机数表法分别分为空白组和模型组,每组16只。选踝关节后外侧后方为穿刺点,将100μL PBS溶液配制成的20 mg/mL的尿酸钠结晶溶液分别注入SD大鼠和Wistar大鼠右足踝关节造模,分别在造模后9 h和12 h各组按随机数字法抽取8只大鼠测量踝关节直径。麻醉后下腔静脉取血,检测白细胞介素-1β(interleukin-1β,IL-1β)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)浓度的变化。结果 Wistar大鼠造模后9 h和12 h模型组关节直径[(0.850±0.201)、(0.235±0.091)mm]较空白组同时间点[(0.080±0.053)、(0.092±0.068)mm]肿胀的差异有统计学意义(P=0.001,P=0.026),SD大鼠造模后9 h和12 h模型组关节直径[(1.128±0.623)、(0.748±0.545)mm]较空白组同时间点[(0.045±0.012)、(0.047±0.015)mm]明显肿胀的差异有统计学意义(P=0.013,P=0.042),2种品系大鼠造模前后关节直径变化差异有统计学意义(P<0.05),但造模后9 h与造模后12 h肿胀程度比较差异均无统计学意义(P>0.05);SD大鼠较Wistar大鼠关节肿胀程度更为明显,差异有统计学意义(P<0.05);且主要炎性因子TNF-α、IL-1β浓度也明显升高,差异有统计学意义(P<0.05)。结论 SD大鼠较Wistar大鼠对尿酸钠晶体导致关节炎症反应更为明显,更适合用于尿酸钠晶体导致的AGA的建立。Objective Although the gouty arthritis model has been applied greatly in practical research,different strains of rats have different responses(reactivity) to sodium urate crystals,which may impact the effect of the model.The aim of the study was to compare the sensitivity of two strains of rats to the model of acute gouty arthritis (AGA) induced by sodium urate crystals through the changes of inflammatory factors,joint diameters and joint pathology.Methods Taking the back of the outside of the ankle as the point of puncture,20 mg/mL sodium urate crystal solution prepared by 100 microliters PBS solution was injected into the right ankles of SD rats and Wistar tats for modeling.Eight rats were selected randomly from each group to measure ankle diameters 9 hours and 12 hours respectively after the modeling.Blood were taken from the inferior vena cava after anesthesia to detect the concentration changes of IL-1 β and TNF-alpha.Results The joints of the model groups of SD rats and Wistar rats were markedly swollen.The variation of the joint diameters before and after the modeling of two strains of rats was meaningful statistically ([0.850 ± 0.201] mm vs [0.080 ± 0.053] mm,[0.235 ± 0.091] mm vs [0.092 ± 0.068] mm,P 〈 0.05).But there was no statistical meaning as to the comparison between the swelling degrees 9 hours and 12 hours after modeling (P 〉 0.05).The swelling degree of SD was more obvious than that of Wistar rats and the variation had statical meaning ([1.128 ±0.623] mm vs [0.045 ± 0.012] mm,[0.748 ± 0.545] mm vs [0.047 ± 0.015] mm,P 〈 0.05),moreover,the concentration of the major inflammatory cytokines of IL-1 beta and TNF-alpha in SD rats was significantly higher in comparison to Wistar rats,which showed statistical meaning (P 〈 0.05).Conclusion Compared with Wistar rats,SD rats showed obvious responses(reactivity) caused by sodium urate crystals.
