边鸡Myf-3基因与生长性状的关联分析

摘  要:本实验采用PCR-SSCP技术检测边鸡生肌决定因子3(Myf-3)的多态性,并分析其对边鸡生长性状的遗传效应。结果表明:在P3位点上发现C576T的突变;在P4位点上发现C1837T和G1846A的突变。P3和P4位点均属于低度多态,其中P3位点处于哈代-温伯格平衡(P>0.05);P4位点偏离了哈代-温伯格平衡(P<0.05)。利用PHASE软件对其进行单倍型分析后结果显示,共产生4种单倍型H1(TCG)、H2(TTA)、H3(CCG)、H4(CTA)。经关联分析发现,单倍型组合H1H1型和H2H2型对所测定的边鸡各周龄体重具有显著或极显著影响。由此推测,边鸡Myf-3基因的多态性对其生长性状有一定影响,但是否可以作为影响边鸡生长性状的遗传标记仍需进一步研究。In this experiment,Bian chicken was used to study the effect of Myf-3 as a candidate gene on chicken growth trait.Five pairs of primers were designed according to the database of chicken Myf-3 gene sequence.PCR-SSCP and DNA sequencing were used to detect the genotypes,and the PHASE software was used as haplotype analysis.Three mutation locus were found.The P3 and P4 locus were low polymorphic,one was agreement with Hardy-Weinberg equilibrium(P>0.05),the other was disagreement with Hardy-Weinberg equilibrium(P<0.05).Using the Phase program,four haplotypes,H1 (TCG),H2 (TTA),H3 (CCG),H4 (CTA)were found.The least square means analysis showed that H1H1 and H2H2 had significant effect on week-age-weight.Polymorphisms of Myf-3 gene had certain effect on growth traits in Bian chicken.However,whether these polymorphisms are important genetic markers,which related with Myf-3 gene and reproduction traits in Bian chicken,are need to be further studied.
