摘  要:目的:建立血清中万古霉素浓度的反相高效液相色谱法的测定方法。方法:以去甲万古霉素为内标,血清样品经10%高氯酸溶液沉淀,采用TC-C18柱(250 mm×4.6 mm,5μm),以甲醇-0.05 mol·L-1磷酸二氢钾(18∶82)为流动相,流速为1.0 ml·min-1,紫外检测波长为230 nm。结果:在1.0~100.0 mg·L-1浓度范围内线性良好(r=0.99989,n=7),回收率高,日内、日间RSD均小于3%,理论塔板数大于6 000,最低检测限为0.12 mg·L-1。结论:本法样品取样量少,处理方法简便,灵敏度高,方法准确稳定,回收率高,适用于临床常规血药浓度监测及相关科研的应用。Objective To develop a method for the determination of vancomycin concentration in human serum by high-perform-ance liquid chromatography (HPLC).Method Norvancomycin was used as internal standard and the sample deposited by 10% per-chloric acid was separated on TC-C18 analysis column (250 mm ±4.6 mm,5 μm)with a mobile phase of mixture of methanol:0.05mol·L^-1 potassium dihydrogen phosphate (18∶82).The flow rate was 1ml/min,the UV detector wavelength was set up 230 nm.Result The calibrated linear plot of vancomycin was within 1.0-100.0 mg/L (r=0.999 89,n=7),the average recoveries in serum were high,intra-day RSD and inter-day RSD were both less than 3%,the theoretical plate number were more than 6 000, the limit of test was 0.12 mg·L^-1 .Conclusion The method is accurate,simple,sensitive and suitable for the study of therapeutic drug monitoring for vancomycin in human serum in clinic and other fields.
