摘 要:对水提麦冬多糖(WPOJ)进行了3种化学修饰,并比较了修饰前后各组分抗肿瘤活性的变化。实验采用氯磺酸-吡啶法制备硫酸化麦冬多糖(S-WPOJ),利用WPOJ与磷酸化试剂和氯乙酸异丙酸反应分别得到磷酸化、羧甲基化麦冬多糖(P-WPOJ、C-WPOJ)。通过红外光谱技术对WPOJ化学修饰前后的结构进行检测,同时利用MTT法对修饰前后麦冬多糖的抗肿瘤效果进行比较分析。结果表明:化学修饰后的WPOJ分别具有硫酸基团、磷酸基团、羧甲基的特征吸收峰,化学修饰后的WPOJ抗肿瘤效果明显有不同程度的提高,其中,羧甲基化麦冬多糖CWPOJ对K562细胞的抑制作用最强。The aim is to chemical modify WPOJ by using three kinds of methods,and to compare the anti-tumor activities of them before and after modification. Tile experiment made S-WPOJ with ehlorosulfonie aeid-pyridine method,the WPOJ was phosphorylated by phosphorylation regent and was earboxymethylated by isopropyl chloroucetate acid. The infrared speetrosocopy was adopted to test the strueture of WPOJ before and after ehemical modification,at the same time,its anti-tumor activities were also analysised by the MTT mothod before and alter modification, and the resuh showed that there are characteristic absorption peak of sulfuric acid, carboxymethyl groups after chemical modification WPOJ respectively,and the modification have obviously improved anti-tumor activities at different levels activities of CWPOJ is the most strongest. phosphate groups, WPOJ of chemical and the anti-tumor activities of CWPOJ is the most strongest.
