摘  要:目的观察冬虫夏草对肾缺血-再灌注(I/R)大鼠肾组织Caspase-12 mRNA和蛋白表达的影响。方法 SD大鼠随机分为假手术组(sham组)、缺血-再灌注组(I/R组)和冬虫夏草(CS)组3组,采用摘除右肾夹闭左肾蒂60 min的方法复制I/R大鼠模型,sham组和I/R组大鼠再灌注后灌服生理盐水(2 mL/d);CS组再灌注后灌服冬虫夏草提取液[5 g/(kg·d)]。结果 sham组肾小球和肾小管结构基本正常,而I/R组可见肾小管坏死改变,各时间点半定量计分均高于sham组(P<0.01),而CS组肾小管病理改善,半定量计分均低于相同时间点I/R组(P<0.01);正常组和sham组肾组织可见少量的Caspase-12 mRNA和蛋白表达,I/R可诱导Caspase-12 mRNA和蛋白表达上调,各时间点I/R组Caspase-12 mRNA和蛋白表达显著高于sham组(P<0.01)。CS组Caspase-12 mRNA和蛋白水平表达显著低于相同时间点I/R组(P<0.01)。sham组有极少量的肾小管上皮细胞凋亡,I/R可诱导肾小管上皮细胞凋亡显著增多,各时间点I/R组凋亡比例均高于sham组(P<0.01),而CS组各时间点的凋亡比例均低于I/R组(P<0.01)。结论冬虫夏草通过下调肾组织中Caspase-12的表达,抑制内质网应激凋亡途径发挥肾保护作用。【Objective】 To observe the effect of Cordyceps Sinensis on the mRNA and protein expression of caspase-12 in rats following renal ischemia and reperfusion. 【Methods】 The SD rats were divided into sham group, ischemia and reperfusion(I/R) group and Cordyceps Sinensis(CS) group randomly. The I/R rats model were established by the means of left renal pedicle was clamped for 60 min of ischemia following excising right kidney. The rats both in sham group and I/R group were treated by intra-gastric administration for physiological saline 2 mL per day. And the rats both in CS group were treated by intra-gastric administration for CS(5 g·kg·d). We observed the kidney pathology changes, renal tubular epithelial cells apoptosis and the mRNA and protein expression of caspase-12 in rats. 【Results】 The structure of glomerulus and renal tubular both in sham group and I/R group is almost completed. However, renal tubular necrosis in the I/R group rats were induced, and the tubulointerstitial semi-quantitative scoring at all the time points in the I/R group rats is significantly higher than that in the sham group rats(P〈0.01). Pathology damage in the CS group rats are ameliorated, the tubulointerstitial semi-quantitative scoring at the same time point in the CS group rats is significantly lower than that in the I/R group rats(P〈0.01). Light mRNA and protein expression of caspase-12 in sham group rats kidney were found, I/R induces significantly increased mRNA and protein expression of caspase-12, which is significantly higher than that in the sham group rats(P〈0.01), the mRNA and protein expression of caspase-12 at the same time point in the CS group rats is significantly lower than that in the I/R group rats(P〈0.01). There is few renal tubular epithelial cells apoptosis in the sham group rats, I/R induces significantly increased renal tubular epithelial cells apoptosis, which is significantly higher than that in the sham group rats(P〈0.01).
