摘 要:目的:研究淫羊藿对早期兔膝骨性关节炎模型软骨的影响.方法:将18只新西兰大白兔随机取6只为正常组,12只制作兔膝骨性关节炎模型;2周后待兔膝骨性关节炎模型建立,均行MRI检查,再将12只造模兔随机分为模型组6只、药物组6只.第15日起每天开始予药物组淫羊藿按质量分数为1 g/kg灌胃,模型组及正常组则给予等量蒸馏水,所有实验兔灌胃4周后,再行MRI检查,测量信噪比(SNR)、T1ρ值,取胫骨平台用实时PCR测定软骨中MMP-3mRNA表达水平,股骨远端行HE和番红-固绿染色.结果:造模2周后,造模组SNR值低于正常组(P<0.05),T1ρ值高于正常组(P<0.01);经治疗后,药物组SNR值高于模型组(P<0.01),T1ρ值低于模型组(P<0.05).模型组较正常组MMP-3mRNA表达水平增加(P<0.05),关节软骨组织结构破坏严重,软骨细胞外基质中蛋白多糖减少;药物组较模型组MMP-3mRNA表达水平下降(P<0.05),软骨结构破坏减轻,蛋白多糖丢失明显减少.结论:淫羊藿可减缓膝骨性关节炎的软骨退变,可能与减少蛋白多糖的丢失、下调软骨中MMP-3的表达有关.Aim:To investigate the effect of epimedium in cartilage of osteoarthritic knee of rabbits in the early stage.Methods:Six rabbits were selected randomly from 1 8 rabbits as normal group,and the rest 1 2 rabbits as control group were made into knee osteoarthritis models.After 2 weeks,all the rabbits were examined by MRI.The 1 2 rabbits with knee osteoarthritis were then randomly divided into 2 groups:model group and epimedium group.The rabbits in epimedium group were received a daily administration of epimedium at a dose of 1 g·kg-1 ·d -1 of body weight from the 1 5 th day.The other rabbits were re-ceived distilled water of the same volume.All the rabbits were examined by MRI and measured the values of signal to noise ratio (SNR)and T1ρagain after 4 weeks treatment.Then all the rabbits were sacri-ficed.Real time PCR was used to detect the expression level of MMP-3 mRNA in cartilage with the tibia platform of right knee.The samples of right femora were stained by HE and safranin-fast green.Results:After two weeks,the SNR value in control group was lower than that in normal group(P&lt;0.05 ).The&amp;nbsp;T1ρvalue in control group was higher than that in normal group (P &lt;0.01 ).Compared with model group,the SNR value was increased(P&lt;0.01 ),the T1ρvalue was decreased (P&lt;0.05)in epimedium group,after 4 weeks treatment.Compared with normal group,the expression level of MMP-3mRNA was increased in model group (P&lt;0.05 )relatively.At the same time,the structure of cartilage was de-stroyed and proteoglycan in cartilage extracellular matrix was decreased in model group.The expression level of MMP-3 mRNA in epimedium group was decreased more significantly compared with that in model group(P&lt;0.05 ).Compared with model group,the proteoglycan was less decreased,and the degree of pathological changes of cartilage showed the lighter destruction in epimedium group.Conclusion:Epime-dium can slow down the degeneration of cartilage of knee osteoarthritis.It might be
