摘 要:选HindⅢ、EcoRⅠ、PstⅠ、SphⅠ、BamHⅠ和BglⅠ 6种限制酶 ,对QAVc_94株和AV_12 7国际标准株的DNA进行酶切图谱比较结果发现 ,QAVc_94株用上述 6种酶酶切得到的片段数分别为 10、3、12、4、6、9条 ,将各片段碱基数相加得出其基因组长 34 7kb ,略高于国内的报道 ,而与Zask等的结果相接近 ,从分子水平上将QAVc_94鉴定为一株EDSV。对照AV_12 7株的酶切结果与文献报道相符。QAVc_94株酶切图谱分析显示 :HindⅢ和EcoRⅠ的酶谱与报道的其他EDSV分离株基本相似 ,HindⅢ的酶谱与国内研究最多的AA_2长春分离株一致 ,EcoRⅠ的酶谱结果与Zask等报道的B8/ 78株相同。而其他 4种限制酶的酶切图谱与已报道的各地分离株相比 ,差异较大 ,如PstⅠ和BglⅠ多出 2~ 5个识别位点。说明鹌鹑源EDSV已与鸡源、鸭源、鹅源EDSV有较大的变异 ,是一株血清型相同而基因型不同的鹌鹑减蛋综合征病毒。The genomes of QAVc_94 and a foreign standard isolate AV_127 were compared through the analysis of 6 different kinds of restriction endonuclease HindⅢ、EcoRⅠ、PstⅠ、SphⅠ、BamHⅠand BglⅠ.The experimental results corroborated that the DNA of QAVc_94 showed 10,3,12,4,6 and 9 fragments respectively.When the length of every fragment was added,the total genome length is 34.7kb,a little more than the data from other isolates in China,but the same as Zask.L's report.The results of RE analysis on AV_127 were conformed to other reports.RE analysis of QAVc_94 also proved that the Restriction patterns of HindⅢ and EcoRⅠ,was generally similar to that of other EDSV isolates known up to now.The QAVc_94' restriction patterns of HindⅢ was consistent with that of AA_2 EDSV isolate from Changchun which has been discussed widely in China,the QAVc_94's mapping of EcoRⅠ was the same as that of B8/78 isolates reported by Zask.But QAVc_94's restriction patterns and the size of the other four Restriction endonuclease were quite different from other isolates.For example,Bgl Ⅰ and PstⅠ had about 2_5 more recognition sites.Therefore,QAVc_94 was far cry from the EDSVs from chicks and ducks et al.This RE analysis testified that QAVc_94 could be the same serotype,but different genome from the other original EDS_76 viruses.
