异体异位发育小鼠睾丸组织中精子蛋白17的表达研究

摘  要:目的采用已建立的未成熟睾丸组织移植模型,研究异体异位生长发育的小鼠睾丸组织中精子蛋白17的表达情况,以评估该移植模型在生殖医学中应用的可行性。方法以1-2天小鼠睾丸组织为供体,7-12周雄性免疫缺陷小鼠为受体进行组织移植;移植8周后收取移植物组织,采用荧光定量PCR法检测移植物中精子蛋白17的基因Spa17的表达,用免疫荧光法和western blot检测精子蛋白17的表达,并与其在正常小鼠睾丸组织的表达进行对比。结果基因检测显示,Spa17在移植物中的表达与其在8周正常小鼠睾丸组织中表达相比有下调;蛋白分析提示,SP17在8周正常小鼠睾丸组织中表达而在小鼠睾丸组织移植物中表达缺失。结论移植物中在精子的精子运动和受精功能中发挥作用的精子蛋白17的基因表达下调及蛋白表达缺失提示,异体异位睾丸组织移植在临床应用的可行性还需进一步评估。Objective To investigate the expression of sperm protein 17 in heterotopic developed testicular tissues for assessing the feasibility of the animal model in the reproductive medicine.Methods Tissue grafting was performed with testes from neonatal Kun-ming mice aging 1-2 days as doners and male immunodeficient mice as recipients.Grafts were collected after 8 weeks of grafting.The expression of Spa17 were determined using real-time quantitative PCR,and SP17 were determined using Immunofluorescence technique and Western blot.For comparison,the expression of Spa17 and SP17 in normal Kun-ming mice were also determined.Results According to the investigation,the expression of gene Spa17 decreased in heterotopic developed testicular tissues in comparision with normal testicular tissues.And ex-pression defects of protein SP17 was found in heterotopic developed testicular tissues.Conclusion The decreased ex-pression of Spa17 and the expression defect of SP17 suggest that more research is need to evaluate the clinic prospect of this animal model.
