摘  要:目的探讨中药材鸡冠花中有效部位止血作用机制。方法鸡冠花的正丁醇部位提取物分别以高、中、低剂量给大鼠连续灌胃给药14d后,剥离腹主动脉,测定大鼠活化部分纤维蛋白原(FIB)、凝血酶时间(TT)、凝血活酶时间(APTT)、凝血酶原时间(PT)各项指标,考察大鼠血液凝集参数的变化情况;给小鼠连续灌胃给药7d后,摘除眼球取血,用ELISA法测定纤溶酶原激活物(t-PA)和纤溶酶原激活物抑制物(PAI-Ⅰ)值。结果鸡冠花正丁醇液能够缩短大鼠APTT、PT值,对二者的影响呈剂量正相关性,对大鼠TT、FIB值的影响不显著;能够降低小鼠的t-PA值,提高PAI-Ⅰ值。结论鸡冠花正丁醇提取物具有良好的止血作用,其作用机制可能与缩短APTT、PT值,抑制纤溶酶原激活物活性,提高纤溶酶原激活物抑制物活性有关。Objective Through animal experiments,to explore hemostatic mechanism of action in effective part of Chinese herbal medicine Celosia cristata L. Methods The extractive of n - butanol parts, continuous intragastric administration to rats 14d with high, medium and lowdose , respectively, and peel the abdominal aorta. To determine fibrinogen (FIB), thrombiu time (3T) , activated partial thromboplastin time (APTT) , prothrombin time (PT) indicators of rat's activation, and investigate changes of blood coagulation parameters; After intragastric administration 7d to rat , the blood was obtained by extirpating their eyes , and measured plasminogen activator ( t - PA) and plasminogen activator inhibitor (PAI - I ) values by ELISA . Results The butanol solution can shorten the APTT, PT value, the impact on both seems to be positive correlation between dose, but the TT, FIB value is not significant ; It can reduce the value of t - PA in mice, and increase PAI -Ⅰ values. Conclusion Butanol extracts has a good hemostatie effect, its mechanism may relative to shorten the APTT, PT values or increase activity of plasminogen activator or inhibit activity of plasminogen activator inhibitor.
