摘 要:目的通过观察隔药灸命门穴对痛经模型大鼠血清前列腺素(PG,Prostaglandin)水平的影响,探讨隔药灸命门穴治疗原发性痛经的作用机制。方法将100只雌性SD大鼠随机分为造模组(80只)和空白组(20只),以缩宫素建立大鼠原发性痛经模型。空白组为正常对照组(A组)并将造模成功的80只大鼠随机分为模型组(B组)、药物组(C组)、针刺组(D组)、隔药灸组(E组)。比较各组大鼠的扭体反应潜伏期和30 min内扭体数并用放免法检测各组大鼠血清PGF2α和PGE2的水平。结果①空白对照组无扭体,模型组扭体次数较多,且有扭体潜伏期,两组比较有非常显著性意义(P<0.01)。与模型组相比,药物组、针刺组以及隔药灸组均能明显延长首次扭体潜伏期,且能减少30 min内扭体反应次数(P<0.01)。药物组、针刺组以及隔药灸组扭体潜伏期比较,差异无统计学意义(P>0.05),但与药物组相比,针刺组及隔药灸组均能明显减少30 min内扭体反应次数(P<0.01);且针刺组与隔药灸组相比,隔药灸组30 min内扭体次数最少(P<0.01)。而且三组比较,隔药灸组扭体抑制率最高。②与空白对照组比较,模型组、药物组、针刺组及隔药灸组大鼠血清PGF2α含量显著增加,PGE2含量显著降低(P<0.01);与模型组比较,药物组、针刺组及隔药灸组PGF2α显著降低,PGE2显著增加(P<0.01);与药物组比较,针刺组及隔药灸组PGF2α显著降低,PGE2显著增加(P<0.01);与针刺组比较,隔药灸组PGF2α降低不明显(P>0.05),但PGE2显著增加(P<0.01)。结论隔药灸治疗原发性痛经与调节PGF2α及PGE2有关,即可降低痛经大鼠血清PGF2α含量,增加血清PGE2含量。Objective To observe the effect of drug- paste interposed moxibustion on the expression of Prostaglandins (PGs) in rats with primary dysmenorrhea, and in order to investigate the mechanism of drug - paste interposed moxibustion on Mingmen (DU4) in treating primary dysmenorrhea. Methods 100 female rats were randomized into Model Group(80 rats) and Control Group (20 rats ), and primary dysmenorrhea model rats were established by extractum leonuri inspissatum (ELI). The blank control group ( A Group), and 80 female mode/rats were divided into four groups, model group ( B Group), medicine group ( C Group ), ac- upuncture group( D Group), drug- paste interposed moxibustion group (E Group). The latency period and the incidence rate of writhing body within 30 minutes were observed in each group and the serum levels of PGF~, and PGE2 were detected by RIA. Results (1)The latency period of writhing body was shorter and the incidence rate of writhing body was more in model group, compared with those of A Group(P 〈0.01 ). The latency period of writhing body was longer and the incidence rate of writhing body was less in C Group, D Group and E Group,compared with those of B Group(P 〈 0.01 ). There was no significant difference between C Group, D Group and E Group in the latency period of writhing body(P 〉 0.05 ). But compared with C Group,the inci- dence rate of writhing body was less in D Group( P 〈 0.01 ) and it was least in E Group (P 〈 0.01 ). Compared with C, D and E Group, the control rate of writhing body was highest in E Group. (~)Compared with A Group, the serum levels of PGF2~ in B, C, D and E Group were obviously ascend (P 〈 0.01 ), while the serum level of PGE2 was obviously descend (P 〈 0.01 ).
