摘  要:目的:观察降脂消斑片对VEGF介导的AS斑块中炎症反应的影响。方法:运用高脂高胆固醇饲料喂养12周造成日本大耳白兔动脉粥样硬化模型,用降脂消斑片灌胃给药治疗4周,采用双抗体夹心酶联免疫吸附法检测血清中ICAM-1、MMP-9的含量;采用免疫组化法分析VEGF、ICAM-1、MMP-9在主动脉粥样斑块中的表达量。结果:与模型组比较,降脂消斑片能明显降低血清中ICAM-1、MMP-9的含量,差异均具有统计学意义(P<0.01);同时也能明显降低VEGF、ICAM-1、MMP-9在主动脉粥样斑块中的面密度,差异均具有统计学意义(P<0.01)。结论:降脂消斑片抑制AS斑块内VEGF的表达,进而降低AS斑块中ICAM-1、MMP-9的表达,抑制斑块内炎症反应,从而达到稳定AS斑块作用,延缓AS的进展,这可能是其抗AS作用的机制之一。Objective: To observe the effects of Jiangzhi Xiaoban pills on inflammatory response in atherosclerosis. Methods: Use the high cholesterol diet to induce the rabbit models for 12 weeks,then fill the Jiangzhi Xiaoban pills to the stomach for four weeks,finally detect the serum levels of ICAM-1,MMP-9 in the rabbits by double antibody sandwich enzyme-linked immunosorbent method and the expression levels of VEGF, ICAM-1,MMP-9 in the atherosclerotic plaques by immunohistochemical staining. Results: Compared with the model group,the Jiangzhi Xiaoban pills could decrease significantly the serum levels of ICAM-1,MMP-9,and the difference was statistically significant( P &lt; 0. 01); meanwhile,the Jiangzhi Xiaoban pills could decrease significantly the surface density of VEGF,ICAM-1,MMP-9 in atherosclerotic plaques,and the difference was statistically significant( P &lt; 0. 01). Conclusion: The Jiangzhi Xiaoban pills inhibit the expression levels of VEGF,then they also decrease the level of MMP-9 and ICAM-1 in the atherosclerotic plaques,and block the inflammatory response. ln these ways,Jiangzhi Xiaoban pills could stabilize the atherosclerotic plaques and delay the progress of atherosclemsis,which may be the mechanisms to resist the atherosclerotic plaques.
