苍附导痰丸加减对多囊卵巢综合征大鼠卵巢形态及性激素水平的影响

摘　　要：目的：研究苍附导痰丸加减对多囊卵巢综合征（PCOS）大鼠卵巢形态及性激素的影响,探讨苍附导痰丸加减治疗PCOS的机制。方法：雌性大鼠随机分成5组,每组大鼠10只。模型组：采用丙酸睾酮＋绒毛膜促性腺激素（HCG）建立PCOS大鼠病理模型;中药高剂量组：造模结束后苍附导痰丸加减24g/kg灌胃;中药低剂量组：造模结束后苍附导痰丸加减6g/kg灌胃;克罗米芬组：造模结束后,第1~14天灌胃给予克罗米芬悬液;空白对照组：未造膜大鼠颈背部皮下注射等量的生理盐水及继予灌胃等量生理盐水。14天后光镜观察卵巢大体形态变化,卵泡发育情况,放射免疫法测定血清卵泡刺激素（FSH）、雄激素（T）、雌二醇（E2）、促黄体生成素（LH）水平。结果：中药组用药后大鼠体质量及卵巢重量减轻,以高剂量组明显;中药高剂量组能显著降低大鼠血清中T、LH水平,增加E2、FSH水平（P〈0.05）。结论：苍附导痰丸加减对丙酸睾酮＋HCG建立的PCOS大鼠有一定的治疗作用,可改善卵巢功能及性激素水平异常。Objective： To investigate the effect of traditional Chinese medicine modified TCM-CFDTW on ovary tissues pathology and improving sex hormone in rat models of PCOS. Methods： Rat models of PCOS were established by the combined administration of testosterone propionate with human chorionic gonadotropin. After the confirmation of successful establishment of PCOS,clomiphene and two different doses TCM-CFDTW decoction 24g / kg,6g / kg were taken respectively by intragastric administration for 14 days. Following the collection of various samples,the rats’ weight and the ovarian weight were dertermined,general morphology of rat ovary and development of follicles were observed under light microscopy. Blood levels of androgen,estradiol, luteinizing hormone（ LH）,follicle stimulating hormone（ FSH） were determined by radioimmunoassay. Results： TCM-CFDTW treatment with higher dosage significantly decreased the rats’ weight and ovarian weight, and circulating levels of androgen,LH. In addition,higher dosage of TCM-CFDTW increased blood levels of estradiol,FSH. It also inceased the follicular wall pellet cellular layer and yellow body quantity. However, TCM-CFDTW treatment at lower dosage did not exhibit a significant efficacy. Conclusion： Modified TCM-CFDTW shows significantly therapeutic efficacy on PCOS in rat models. This effect of TCM-CFDTW mainly depends on improving PCO situation and improving sex hormone.
