养阴润燥败毒合剂治疗放射性肠炎大鼠作用机制的初探

摘  要:为探讨养阴润燥败毒合剂治疗放射性肠炎大鼠的作用机制,本实验采用直线加速器局部单次大剂量腹盆腔照射法建立放射性肠炎的大鼠模型,将36只造模后的大鼠随机分为实验组和对照组,各18只,实验组以养阴润燥败毒合剂灌胃,对照组以庆大霉素加地塞米松的混合液灌胃,连续7d;分别于灌胃治疗第3天、第5天和第7天用脱颈法处死大鼠;采用酶联免疫吸附测定(ELISA)法检测大鼠血清白细胞介素(IL)-1β、IL-6和肿瘤坏死因子-α(TNF—α)3种炎症因子的含量,采用反转录聚合酶链反应(RT—PCR)法检测大鼠肠道组织中核因子-κB(NF-κB)mRNA和过氧化物酶增殖体激活受体-γ(PPAR-γ)mR—NA的表达水平。结果显示,随着灌胃治疗时间的延长,两组大鼠血清IL-1β、IL-6、TNF—α的含量及肠道组织NF-κB mRNA的表达水平逐渐下降,而PPAR-γmRNA的表达水平逐渐升高。其中实验组大鼠血清IL-1β、IL-6含量及肠道组织NF-κBmRNA、PPAR-γmRNA表达水平变化程度更为明显,P〈0.05;两组大鼠血清TNF-α含量变化程度比较差异无统计学意义,P〉0.05。结果表明,养阴润燥败毒合剂治疗放射性肠炎可能是通过抑制炎症因子的表达实现的。This study was to probe into the influence mechanism of the mixture for nourishing Yin to moisten dryness and clearing away toxic material in the treatment radioactive enteritis in rats. In the study, by using linear accelerator to irradiate abdominopelvic cavity locally and at single-time large-dosage so as to establish radioactive enteritis model of rats, to divided the 36 models of rats into experimental group and control group( n = 18, each one), to pour the mixture into the stomach of rats of experimental group,while to pour the mixture solution of Gentamycin and Dexamethasone into the stomach of rats of control group, for 7 days in both groups;on the 3rd,5th and 7th days after above treatment the rats were sacrificed by dislocation of neck; by using enzyme-linked immunosorbent assay(ELISA) to detect the level of interleukin (IL)-113, IL-6 and tumor necrosis factor-α(TNF-α) in rats’ serum; by using reverse transcription polymerase chain reaction(RT-PCR) to detect the expression levels of muclear factor-κB(NF-κB) mRNA and peroxisome proliferator activated receptor-γ(PPAR-γ) mRNA in rats' intestinal tissue. As results, with treatment time was lengthened the expression levels of IL-1β,IL-6 and TNF-α,as well as the levels of NF-κB mRNA in rats' intestional tissue gradually declined, but PPAR-γ mRNA expresstion level lifted, either experimental and control group;Also,compared with control group the expression levels of IL-1β, IL-6, and the expression levels of NF-κB mRNA and PPAR-γ mRNA in experimental group had more significant changes( P 〈0.05), but in TNF-α level change there was no statistical difference between both groups( P 〉0.05). Results show that the mixture for nourishing Yin to moisten dryness and clearing away toxic material in the treatment of radioactive enteritis could obtain therapeutic effect through inhibiting the expression of inflammatory factors.
