摘  要:目的:探讨金雀异黄素(Gen)对氧化损伤的人晶状体上皮细胞(HLEC)凋亡有无抑制作用。方法:本研究在终浓度3×10-4mol/L H2O2培养液中复制HLEC凋亡模型,并以雌二醇(E2)作为阳性对照,采用流式细胞仪(FCM)检测HLEC凋亡率和线粒体膜电位(ΔΨM)的变化。结果:FCM结果显示:H2O2组HLEC凋亡率显著高于空白对照组;E2和Gen组凋亡率均低于H2O2组(P<0.01)。H2O2组线粒体膜电位比对照组显著下降;E2组和Gen组的线粒体膜电位均比H2O2组升高(P<0.01)。结论:Gen可减轻实验性氧化损伤的HLEC凋亡程度。Objective: To investigate if Genistein( Gen) inhibits the apoptosis of human lens epithelial cell( HLEC) resulted from the experimental oxidative injury. Methods: HLECs were incubated with 3 × 10^- 4mol / L H2O2. And make estradiol( E2) as a positive control. HLECs apoptosis and apoptosis rate were measured by FCM. Results: Apoptosis rate in H2O2group was significandy higher than that in control group,E2and Gen group. The mitochondria transmembrane potential in H2O2group reduced obviously,comparing to control group,E2and Gen group. Conclusion: Gen may decrease the apoptosis of HLEC resulted from the experimental oxidative injury,providing scientific evidence for pursue effective medicine to prevent and treat cataract.
