摘　　要：目的　为建立出血性大肠杆菌O15 7:H7DNA文库 ,筛选致病岛LEE。方法　提取EHECO15 7:H7EDL933染色体 ,Xmal部分酶切并与pJB8X连接、包装 ,感染E .coliDH5α,鉴定库容。结果　库容约 70 0克隆子 ,大于 6 0 8个理论值 ,构建成功。结论　成功构建了出血性大肠杆菌O15 7:H7DNA文库 ,为目的片段筛选打下良好基础。Objectives To construct EHEC O157:H7 DNA bank for th screening of pathogenicity island LEE.Methods Chromosomal DNA of Enterohaemorrhagic E.Coli O157:H7 was extracted and digested with Xmal.Then it was ligated to pJB8X and packed with λ phages.The products of package were used to infect E.coli DH5 α and the capacity of DNA bank was identified.Results The content of the bank was about 700 clones,which was larger than that of the theoretical value.Conclusion The bank has been constructed successfully for screening any fragment you want.
