摘 要:目的:研究不同程度热量限制(calorie restriction,CR)对卵巢储备功能的影响。方法:取8周健康雌性C57BL/6小鼠30只,随机分为自由饮食组(adlibitum,AL)、25%CR组和45%CR组,8周后评价小鼠的卵巢储备功能。通过对卵巢连续切片进行HE染色后光镜下卵泡计数和Real-Time PCR技术检测小鼠卵巢抗苗勒氏管激素(AMH)mRNA的表达改变检测卵泡储备功能,并利用Real-Time PCR技术检测小鼠卵巢FOXO3a和CYP11A mRNA的表达。结果:AL、25%CR和45%CR小鼠体重均呈现波动增长的趋势。卵泡计数结果显示,AL组小鼠卵巢始基卵泡数(352.0±100.1)个,低于25%CR组小鼠卵巢始基卵泡数(1 167.0±294.8)个(P<0.05)。25%CR组小鼠卵巢AMH的mRNA的相对表达量高于AL组(P<0.05),与卵巢计数始基卵泡结果一致。25%CR组始基卵泡的比例为36.59%,45%CR组始基卵泡的比例为30.88%,均显著高于AL组始基卵泡的比例(15.97%)。检测各组小鼠卵巢内FOXO3a和CYP11A的mRNA相对表达量,可见与AL组小鼠相比25%CR组小鼠卵巢CYP11A表达明显上调,45%CR组小鼠卵巢FOXO3a表达明显上调。结论:8周健康雌性C57BL/6小鼠以25%CR进行为期8周的模型构建,可以在不影响生长发育的前提下达到保护卵巢储备功能的目的。Objective: To research the effects of caloric restriction (CR) in varying degrees on ovarian reserve function. Methods: Thirty healthy female C57BL/6 mice of 8 weeks old were randomly divided into ad libitum (AL) group, 25% CR group and 45% CR group. Their ovarian reserve functions were evaluated after 8 weeks. The serial sections of ovaries were stained by HE staining, follicular count under light microscopy was conducted, RT- PCR was used to detect the expression of ovarian anti -Mtlllerian hormone (AMH) mRNA of mice, ovarian reserve function was evaluated, RT -PCR was also used to detect the expressions of ovarian FOXO3a and CYP11A mRNA of mice. Results: Body weights of mice in AL group, 25% CR group and 45% CR group showed fluctuational increasing trends; the results of follicular count showed that the number of ovarian primordial follicle in AL group was (352. 0 ± 100. 1 ), which was statistically significantly lower than that in 25% CR group ( 1 167.0 ±294. 8 ) ( P 〈 0. 05 ) ; the relative expression level of AMH mRNA in 25% CR group was statistically significantly higher than that in AL group ( P 〈 0. 05 ), which was consistent with the number of ovarian primordial follicle by follicular count; the proportions of ovarian primordial follicle in 25 % CR group and 45 % CR group were 36. 59% and 30. 88 %, respectively, which were significantly higher than that in AL group (15.97%) ; compared with AL group, the expression of CYPllA mRNA in 25% CR group increased significantly, the expression of FOXO3a in 45% CR group increased significantly. Conclusion: Among the healthy female C57BL/6 mice of 8 weeks old, 25% calorie restriction for 8 weeks can achieve the goal of protecting ovarian reserve function on the premise of not affecting growth and development of these mice.
