摘 要:目的:探讨健胎液对胚胎着床障碍小鼠着床期子宫炎性细胞因子IL-1β及IL-1RⅠ的影响。方法:建立胚胎着床障碍的动物模型,分为三组,观察各组着床率和着床点数,妊娠第5天分别检测各组小鼠子宫内膜IL-1β的蛋白含量,以及子宫内膜IL-1β、IL-1RⅠ的mRNA水平。结果:模型组胚胎着床数、着床位点数与正常组比较显著降低(P<0.01);中药组胚胎着床数、着床位点数与模型组比较明显升高(P<0.05),但仍低于正常组(P<0.05)。模型组子宫内膜IL-1β蛋白量同正常组、中药组比较明显升高,中药组IL-1β蛋白量同正常组比较无显著性差异。模型组IL-1β、IL-1RⅠ的mRNA表达水平同正常组比较显著升高,中药组IL-1RⅠ的mRNA表达水平同模型组比较明显降低,但仍比正常组水平高。结论:围着床期子宫内膜IL-1β、IL-1RⅠ参与胚胎着床的病理生理过程,健胎液可能通过调节IL-1表达,改善子宫内膜微环境,提高子宫内膜胚胎种植容受性,促进着床。Objective:We discussed the effect of IL-1β,IL-1R of mouse models with embryo implantation dysfunction during the peri-implantation period treated with Jiantai liquid. Methods:The embryo implantation dysfunction model was established. Ninety cases were divided into 3 groups. We observed the implantation rate and the number of implantation sites of each group. The protein levels of IL-1β in the endometrium were examined. The mRNA levels of IL-1β,IL-1R in the endometrium were examined. Results:It was demonstrated that the implantation rate and the number of implantation sites of model group were signifi-cantly lower than that of control group(P〈0.01). The implantation rate and the number of implantation sites of CHM group were higher than those of model group,but still lower than that of control group(P〈0.05). The concentration of IL-1β in model group was significant higher than that of control group and CHM group. The concentration of IL-1β and IL-1RⅠmRNA in model group were significant higher than that of control group. The concentration of IL-1 RⅠ mRNA in CHM group was significant lower than model group , but still higher than that of control group. Conclusion:It is implicated that the possible mechanism of CHM might be to regulate the expression and transcription of IL-1β,IL-1R and improve the microenvironment of endometrium,as well as increase its reception to the embryo.
