摘 要:[目的]研究不同冻融条件对破壁提取巢湖水华新鲜蓝藻中藻蓝蛋白的影响,为后续提取纯化试剂级藻蓝蛋白提供支持。[方法]在各种条件下,冻融破壁后,利用粗提液中藻蓝蛋白即PC纯度和得率为评价指标,研究不同冻融次数、不同含水率和不同冻融介质对藻蓝蛋白提取效果的影响。[结果]粗提液中PC纯度随着冻融次数的增加而依次减小,PC得率随着冻融次数的增加呈现下降的趋势;PC纯度和得率随着含水量的增大呈现先持平后降低的趋势;在不同冻融介质的试验中,通过比较得出最优的冻融介质为浓度0.0025mol/L的PBS缓冲溶液。[结论]对于冻融储存时间较长的巢湖新鲜蓝藻,最优的试验条件是选用浓度为0.0025mol/L的PBS缓冲液作为冻融介质,在含水率为96.O%~97.5%区间冻融次数仅需1次,即可得到最优的PC的纯度和得率。[ Objective ] To study effects of different freezing and thawing conditions on extraction of the phycocyanin from fresh blue algae, and provide support for extlaction of phycocyanin reach the reagent grade level. [ Method ] Taking repeated freeze-thaw cycles, the gradient moisture content, the different medium of freezing and thawing experiment were compared based on the phycocyanin purity value and recovery. [Result] Results showed that with the increase of freezing and thawing, the purity values were decreased in turn, and the numerical of recovery were ups and downs. The experiments of the gradient moisture content showed that the purity value and yield value were presented from held the line to decline. The phycocyanin purity and recovery showed that the buffer solution of PBS is better than others, and so is recommen- ded as the optimal medium for the experiments of freeze thawing cycle. [ Conclusion] Based on the particularity of preservation conditions for the unnecessarily long of storage, freeze thawing one time can be used as a selection. The result showed that the optional medium is PBS buffer solution, the concentration is 0. 002 5 mol/L, and the moisture content of blue alga from 96% to 97.5% is better than others, so it is recommended as a common standard for phycocyanin extraction from blooms water samples in Lake Chaohu.
